人参皂苷Rg3对光老化成纤维细胞中转录因子E2相关因子2与超氧化物歧化酶表达的影响 被引量:2

Effect of Ginsenoside Rg3 on the expression of nuclear factor erythroid 2 related factor 2 and superoxide dismutase in photoaging fibroblasts

摘要 目的探讨人参皂苷Rg3对中波紫外线(UVB)致体外培养人成纤维细胞光老化的影响。方法体外培养人成纤维细胞,用UVB照射建立光老化模型。将细胞分为正常组、模型组、对照组和实验组。正常组给予37℃、5%CO2条件下常规培养;模型组给予UVB一次性照射,照射剂量为20 mJ·cm^(-2),照射时间为222.22 s;对照组给予含40μg·mL^(-1)人参皂苷Rg3的培养基进行培养;实验组给予含40μg·mL^(-1)人参皂苷Rg3的培养基培养,并用UVB一次性照射,剂量为20 mJ·cm^(-2)。培养48 h后,用荧光探针法检测细胞活性氧(ROS)水平,用流式细胞仪检测细胞周期,用实时荧光定量聚合酶链反应检测转录因子E2相关因子2(Nrf2)和超氧化物歧化酶(SOD) mRNA的表达水平。结果正常组、模型组、对照组和实验组的ROS分别为22.42±3.26,48.38±4.79,19.52±3.42和32.72±4.74,G1/G0期百分比分别为(62.85±2.25)%,(70.67±1.92)%,(58.58±1.93)%和(60.30±2.31)%,S期分别为(15.27±1.03)%,(8.28±0.63)%,(18.52±1.28)%和(17.23±1.36)%,Nrf2 mRNA相对表达量分别为1.00±0,1.18±0.06,1.04±0.03和1.26±0.04,SOD mRNA相对表达量分别为1.00±0,1.21±0.05,1.33±0.06和1.34±0.08。模型组、对照组和实验组的上述指标与正常组比较,差异均有统计学意义(均P <0.05)。结论人参皂苷Rg3可以减轻UVB致体外培养人成纤维细胞光损伤,主要机制可能是通过Kelch样环氧氯丙烷相关蛋白1-转录因子E2相关因子2-抗氧化反应元件信号通路调控SOD产生,刺激细胞活性,清除ROS。 Objective To investigate the effect and mechanism of Ginsenoside Rg3 on the photoaging fibroblasts of human induced by ultraviolet B(UVB) in vitro.Methods Human fibroblasts were cultured in vitro and the photoaging model was established by UVB irradiation.The cells were divided into normal,model,control and experimental groups.Normal group was given normal culture with 37℃,5% CO2 Model group was given one-time irradiation of UVB with irradiation dose of 20 mJ·cm^(-2),and irradiation time of 222.22 s.Control group was given the medium to interfere with the culture.Experimental group was cultured with a medium containing 40 μg·mL^(-1)Ginsenoside Rg3,and one-time irradiation of UVB with irradiation dose of 20 mJ·cm^(-2).After 48 hours of culture reactive oxygen species(ROS) levels were detected by fluorescence,cell cycle was detected by flow cytometry,mRNA expression of nuclear.factor erythroid 2 related.factor(Nrf2) and super oxide dismutase(SOD) were detected by real-time fluorescence quantitative polymerase chain reaction.Results The ROS levels of normal group,model group,control group and experimental group were 22.42±3.26,48.38±4.79,19.52±3.42 and 32.72±4.74,the percentage of cells in G1/G0 phase were(62.85±2.25) %,(70.67±1.92) %,(58.58±1.93) % and(60.30±2.31) %,the percentages of S phase cells were(15.27±1.03) %,(8.28±0.63) %,(18.52±1.28) % and(17.23±1.36)%,Nrf2 mRNA expression levels were 1.00±0,1.18±0.06,1.04±0.03 and 1.26±0.04,SOD mRNA expression levels were 1.00±0,1.21±0.05,1.33±0.06 and 1.34±0.08.There were statistically significant differences between model group,control group and experimental group and normal group(all P <0.05).Conclusion Ginsenoside Rg3 can reduce UVB induced photodamage of cultured human fibroblasts,and the main mechanism may be through the kelch like epichlorohydrin related protein 1-nuclear factor erythroid 2 related factor 2-antioxidant response element signaling pathway to regulate SOD production,stimulate cell activity,and eliminate ROS.
