摘要
目的检测脑胶质瘤组织中反义长链非编码RNA(LncRNA)-antisense transense of FAS(FAS-AS1)的表达情况,并探讨其与患者临床病理参数和预后的关系。方法收集2015年1月至2016年12月在沧州市中心医院行镜下手术全切除的脑胶质瘤患者58例为研究对象,选取同期因颅脑外伤行手术切除的患者42例为对照组,分别取脑胶质瘤患者及颅脑外伤患者手术切除后的脑胶质瘤组织及正常脑组织,采用实时定量聚合酶链反应(qRT-PCR)法检测两组脑组织中LncRNA FAS-AS1的表达情况。以脑胶质瘤组织中LncRNA FAS-AS1表达水平的均值为界,将脑胶质瘤分为高表达组(32例)与低表达组(26例),比较不同LncRNA FAS-AS1表达组患者的临床病理参数。并对脑胶质瘤患者进行2年随访,分析LncRNA FAS-AS1表达与患者预后的关系。结果脑胶质瘤组织中LncRNA FAS-AS1的表达水平显著低于正常脑组织[(1.01±0.32)vs.(2.05±0.21),t=7.544,P<0.05]。LncRNA FAS-AS1低表达组患者肿瘤直径、病理分级、瘤周脑水肿(PTBE)程度与高表达组差异均有统计学意义(P<0.05)。随访2年,58例脑胶质瘤患者中有28例死亡,其中LncRNA FAS-AS1低表达组患者2年生存率显著低于高表达组(30.77%vs.68.75%,Log-rankχ2=7.674,P<0.05)。多因素Cox回归分析结果显示,LncRNA FAS-AS1低表达、病理分级Ⅲ~Ⅳ级、重度PIBE是影响脑胶质瘤患者预后的独立危险因素。结论脑胶质瘤组织中LncRNA FAS-AS1表达下调,其可能成为判断脑胶质瘤患者预后的标记物之一。
Objective To detect the expression of antisense long non-coding RNA(LncRNA)-antisense transense of FAS(FAS-AS1)in brain glioma tissues and explore its relationships with clinical pathological parameters and prognosis.Methods From January 2015 to December 2016,58 patients with brain glioma underwent total resection under endoscope in Cangzhou Central Hospital were selected as the study subjects,and 42 patients underwent surgical resection due to craniocerebral trauma in the same period were selected as the control group.Brain glioma tissues and normal brain tissues were taken from patients with brain glioma and patients with traumatic brain injury after surgical resection.Real-time quantitative polymerase chain reaction(qRT-PCR)was used to detect the expression of LncRNA FAS-AS1 in brain glioma and normal brain tissues.With the mean expression level of LncRNA FAS-AS1 in glioma tissue as the boundary,glioma tissues were divided into high expression group(32 cases)and low expression group(26 cases),the clinicopathological parameters of different LncRNA FAS-AS1 expression groups were compared.The patients with brain glioma were followed up for 2 years,and the relationship between the expression of LncRNA FAS-AS1 and the prognosis of patients with glioma was analyzed.Results The expression level of LncRNA FAS-AS1 in glioma tissue was significantly lower than that in normal brain tissue([1.01±0.32]vs.[2.05±0.21],t=7.544,P<0.05).There were significant differences in tumor diameter,pathological grade and degree of peritumoral brain edema(PTBE)between the high expression group of LncRNA FAS-AS1 and the low expression group(P<0.05).During the 2-year follow-up,28 of 58 glioma patients died,and the 2-year survival rate of patients in low expression group of LncRNA FAS-AS1 was significantly lower than that of patients in high expression group of LncRNA FAS-AS1(30.77%vs.68.75%,Log-rankχ2=7.674,P<0.05).Cox model multivariate analysis showed that lower expression level of LncRNA FAS-AS1,pathological grade(Ⅲ-Ⅳ)and severe d
