摘要
目的探讨骨形成蛋白(BMP)-6和BMP-9在糖尿病合并骨折时的表达改变及其与骨折愈合的关系。方法将44只6周龄雄性SD大鼠采用Excel简单随机化分法分为糖尿病组(DM,n=22)和非糖尿病组(ND,n=22)。DM组腹腔注射链脲佐菌素枸橼酸溶液(STZ)50 mg/kg,ND组注射等体重比枸橼酸溶液。2周后于两组动物左股骨制造横断骨折。骨折后的第2、4、8周行影像学和组织形态学骨折愈合评估。通过机械负荷评估骨痂的强度。采用免疫印迹法(Western blot)定量分析骨组织中BMP-6和BMP-9的表达变化。影像学结果以Image J软件进行图像分析。实验数据以SPSS 13软件进行t检验分析。结果骨折后4周糖尿病组骨痂显著小于非糖尿病组(DM组1.64±0.14,ND组2.14±0.18,t=5.499,P<0.05,n=8);骨痂内钙化比例显著低于非糖尿病组[DM组(53.40±6.49)%,ND组(60.61±2.13)%,t=1.976,P<0.05,n=8];骨痂机械强度显著低于非糖尿病组[DM组(161.85±38.33) kPa,ND组(217.49±39.63) kPa,t=2.135,P<0.05,n=6]。相对分子质量为35×10^(3)的BMP-6表达在DM组骨折股骨中表达显著低于ND组骨折股骨。分析较大相对分子质量BMP-6条带与35×10^(3) BMP-6的变化关系,结果提示,35×10^(3) BMP-6的显著下调可能与BMP-6的成熟过程受到糖尿病病理因素抑制有关。BMP-9的表达与糖尿病和骨折无关。结论糖尿病骨折愈合过程受损与BMP-6 35×10^(3)片段表达下调相关。而BMP-6的成熟过程障碍可能是导致上述差异的重要原因。
Objective the Aim of this study was to investigate the effect of diabetes on fracture healing procedure in a rat model.Methods Forty-four adult male SD rats were randomly assigned into diabetes mellitus group(DM,n=22)and non-diabetic group(ND,n=22),via Excel simple randomization.Diabetes was induced by intraperitoneal injection of streptozotocin(STZ,50 mg/kg),while the non-diabetic group received citric acid buffer.Two weeks after induction of diabetes,transverse fracture of the left femur was induced in the rats of both groups.At the end of 2nd,4th and 8th post-fracture week,the radiological,the mechanical and the histomorphology evaluations on were conducted.Expression of bone morphogenetic protein(BMP)-6 and BMP-9 was quantified with Western blotting assay.Results At the end of 4th week of fracture,significantly smaller sizes of callus were observed in the diabetes group(DM 1.64±0.14,ND 2.14±0.18,t=5.499,P<0.05,n=8).Significantly lower calcification ratios were detected in the diabetes group,compared to that in the non-diabetes group[DM(53.40±6.49)%,ND(60.61±2.13)%,t=1.976,P<0.05,n=8].Significantly reduced mechanical endurance in the diabetes group was observed,compared to that of the non-diabetes group[DM(161.85±38.33)kPa,ND(217.49±39.63)kPa,t=2.135,P<0.05,n=6].Furthermore,the expression of 35×10^(3) BMP-6 was significantly lower in the diabetic rats compared to their non-diabetic counterparts.The analysis of all BMP-6 strips indicated that the decrease in 35×10^(3) BMP-6 may be caused by inhibition of the maturation the BMP-6 with a molecular weight of 35×10^(3),which may be caused by the diabetic pathology.BMP-9 was found neither related to diabetes nor fracture healing.Conclusion The findings of this study suggest that the impaired fracture healing procedure in diabetic rats was associated with the decrease of 35×10^(3) BMP-6.
