连翘苷对肺炎克雷伯菌感染的肺炎小鼠的影响 被引量:11

Effect of forsythin on Klebsiella pneumonia infected mice

摘要 目的探讨连翘苷对肺炎克雷伯菌(Kp)感染的肺炎小鼠的影响。方法经口插入气管,按0.28 mL·kg^(-1)的剂量注射2×108 CFU·mL^(-1)Kp菌液构建Kp感染的肺炎小鼠模型,并随机分为模型组和低、中、高剂量实验组,每组10只;空白组10只经口插入气管注射等体积0.9%NaCl。低、中、高剂量实验组分别按50,100和200 mg·kg^(-1)的剂量给予50 mg·L^(-1)连翘苷;空白组和模型组均灌胃给予等体积0.9%NaCl。5组小鼠每天给药1次,连续给药7 d。用流式细胞仪检测辅助性T细胞17(Th17)和调节性T细胞(Treg)细胞含量,用蛋白质印迹法检测肺组织中磷酸化信号转导与转录激活因子5(p-STAT5)和叉头蛋白P3(Foxp3)蛋白的表达水平。结果低、高剂量实验组和模型组、空白组的Th17分别为(31.56±4.62)%,(16.66±3.44)%,(54.19±6.16)%和(16.16±3.15)%,Treg分别为(16.48±2.11)%,(22.44±2.94)%,(11.36±2.49)%和(34.15±4.13)%,STAT5蛋白相对表达量分别为1.26±0.23,1.21±0.23,1.21±0.22和1.23±0.15,p-STAT5/STAT5分别为0.13±0.02,0.26±0.04,0.08±0.01和0.31±0.03,Foxp3蛋白相对表达量分别为0.17±0.02,0.74±0.08,0.06±0.01和0.83±0.08。与模型组相比,低、高剂量实验组血液中Th17水平均显著降低,Treg水平和肺组织中p-STAT5、Foxp3蛋白水平均显著升高,差异均有统计学意义(均P<0.05)。结论连翘苷在Kp感染的肺炎小鼠中可激活STAT5/Foxp3通路,从而促进Th17/Treg细胞平衡,达到缓解疾病的作用。 Objective To investigate the effect of forsythin on Klebsiella pneumonia(Kp)infected mice.Methods The mouse model of pneumonia caused by Kp infection was established by injecting 2×108CFU·m L^(-1)Kp solution into the trachea at the dose of 0.28 m L·kg^(-1).The model mice were randomly divided into model group and experimental-L,-M,-H groups with 10 cases per group.And 10mice in the blank group were injected with the same volume of 0.9%Na Cl through the trachea.Experimental-L,-M,-H groups were given 50 mg·L^(-1)forsythin at the dose of 50,100 and 200 mg·kg^(-1),respectively.Five groups were treated once a day for 7 days.The contents of helper T cell 17(Th17)and regulatory T cell(Treg)were detected by flow cytometry.Western blotting was used to detect the expressions of phosphorylated signal transducer,activator of transcription5(p-STAT5)and forkhead protein P3(Foxp3)in lung tissue.Results The levels of Th17 in experimental-L,-H groups,model group and blank group were(31.56±4.62)%,(16.66±3.44)%,(54.19±6.16)%and(16.16±3.15)%,the levels of Treg were(16.48±2.11)%,(22.44±2.94)%,(11.36±2.49)%and(34.15±4.13)%,the relative expressions of STAT5 protein were1.26±0.23,1.21±0.23,1.21±0.22 and 1.23±0.15,the p-STAT5/STAT5 were 0.13±0.02,0.26±0.04,0.08±0.01 and 0.31±0.03,the relative expressions of Foxp3 protein were 0.17±0.02,0.74±0.08,0.06±0.01and 0.83±0.08.Comparing with the model group,the levels of Th17 in the blood of experimental-L and-H groups decreased significantly,and the levels of Treg in the blood and the levels of p-STAT5 and Foxp3 protein in lung tissue increased significantly(all P<0.05).Conclusion Forsythin can activate STAT5/Foxp3 pathway in Kpinfected pneumonia mice,thus promoting the balance of Th17/Treg cells and relieving the disease.
