microRNA-195-5p在妊娠糖尿病患者血浆中的表达及其对细胞增殖和凋亡的影响 被引量:7

Expression of microRNA-195-5p in plasma of patients with gestational diabetes mellitus and its effect on cell proliferation and apoptosis

摘要 目的探讨微小RNA-195-5p(miR-195-5p)在正常妊娠及妊娠糖尿病患者血浆中的表达差异,并探究其对高糖诱导的人绒毛膜滋养层细胞HTR-8/SVneo细胞增殖和凋亡的影响。方法选取剖宫产分娩且患妊娠糖尿病的产妇34例作为糖尿病组,以及同期本院进行剖宫产的正常产妇34例作为健康组,用实时荧光定量逆转录聚合酶链反应(qRT-PCR)法检测糖尿病组和健康组血浆中miR-195-5p的相对表达水平。将HTR-8/SVneo细胞随机分为正常组、高糖组(给予1000 mg·L^(-1)葡萄糖处理)、实验组1(转染抑制剂阴性对照后,给予高糖组条件培养)、实验组2(转染miR-195-5p抑制剂后,给予高糖组条件培养)。用qRT-PCR法检测HTR-8/SVneo细胞中miR-195-5p的相对表达水平;用噻唑蓝(MTT)法测定细胞的增殖;用流式细胞术检测细胞凋亡情况。结果糖尿病组和健康组血浆中miR-195-5p的相对表达水平分别为1.63±0.71和0.98±0.46,糖尿病组血浆中miR-195-5p的表达水平显著高于健康组(P<0.05)。正常组、高糖组、实验组1、实验组2的HTR-8/SVneo细胞中miR-195-5p的相对表达水平分别为1.03±0.06,1.79±0.19,1.82±0.12,1.26±0.08;细胞增殖率分别为(97.24±4.13)%,(53.51±6.78)%,(47.39±3.25)%,(71.56±6.49)%;凋亡率分别为(5.36±0.42)%,(14.79±1.25)%,(15.38±0.76)%,(8.64±0.56)%。高糖组与正常组比较、实验组2与实验组1比较,差异均有统计学意义(均P<0.05)。结论miR-195-5p在妊娠糖尿病患者血浆中高表达,下调miR-195-5p的表达可以促进高糖诱导的HTR-8/SVneo细胞增殖,并抑制细胞凋亡。 Objective To investigate the difference in the expression of microRNA-195-5 p(miR-195-5 p)in the plasma of normal pregnant and patients with gestational diabetes mellitus,and to explore the effects of miR-195-5 p on the proliferation and apoptosis of human choriotrophoblast HTR-8/SVneo cells induced by high glucose.Methods Thirty-four pregnant women with gestational diabetes who underwent cesarean section were selected as diabetes group,and 34 normal pregnant women who underwent cesarean section during the same period were selected as healthy group.Real-time fluorescence quantitative polymerase chain reaction(qRT-PCR)was used to detect the relative expression level of miR-195-5p in the plasma of diabetes group and healthy group.HTR-8/SVneo cells were randomly divided into normal group,high glucose group(1000 mg·L^(-1) glucose treatment),experimental group 1(transfection with inhibitor negative control,followed by 1000 mg·L^(-1) glucose treatment),and experimental group 2(transfection with miR-195-5 p inhibitor,followed by 1000 mg·L^(-1) glucose treatment).The relative expression level of miR-195-5 p in HTR-8/SVneo cells was detected by qRT-PCR.The methyl thiazolyl tetrazolium(MTT)assay was used to measure cell proliferation.Cell apoptosis was detected by flow cytometry.Results The relative expression levels of miR-195-5 p in the plasma of diabetes group and healthy group were 1.63±0.71 and 0.98±0.46,the expression level of miR-195-5 p in the plasma of diabetes group was significantly higher than that of healthy group(P<0.05).The relative expression levels of miR-195-5 pin HTR-8/SVneo cells of normal group,high glucose group,experimental group 1 and experimental group 2 were1.03±0.06,1.79±0.19,1.82±0.12 and 1.26±0.08,the proliferation rates were(97.24±4.13)%,(53.51±6.78)%,(47.39±3.25)%and(71.56±6.49)%,the apoptosis rates were(5.36±0.42)%,(14.79±1.25)%,(15.38±0.76)%and(8.64±0.56)%.The differences between high glucose group and normal group,as well as between experimental group 2 and experimental gr
