摘要
目的探讨自然杀伤(nature killer,NK)细胞在肝细胞性肝癌合并细粒棘球蚴(Echinococcus granulosus,Eg)感染的裸鼠移植瘤中的表达情况。方法将40只裸鼠随机分为单独移植组和联合移植组,单独移植组将5×10^(6)个HepG2细胞接种于裸鼠皮下,而联合移植组将5×10^(6)个HepG2细胞和2000个Eg原头蚴(protoscolices,PSCs)联合接种于裸鼠皮下。自接种第25天起每4 d测量皮下移植瘤的体积,共5次,接种37 d获取皮下移植瘤。通过苏木素伊红(hematoxylin and eosin,HE)染色观察皮下移植瘤基本病理变化。免疫组织化学染色和实时荧光定量多聚核苷酸链式反应(real-time quantitative polymerase chain reaction,qRT-PCR)法检测CD16、CD56、NKG2D在皮下移植瘤的表达情况。结果与单独移植组相比,不同接种时间联合移植组中移植瘤的体积均明显增大,差异具有统计学意义(P<0.05)。单独移植组中细胞形态和体积不均一,细胞质嗜碱,核质比例增加,有丝分裂活性增强,部分细胞出现脂肪变性、凝固性坏死和空泡。联合移植组中细胞异质性更为明显并可见典型的Eg PSCs结构。免疫组织化学染色结果显示,CD16、CD56、NKG2D阳性细胞主要在皮下移植瘤细胞质中表达,呈棕褐色。定量结果显示,在联合移植组中CD16、CD56、NKG2D表达量明显高于单独移植组,差异具有统计学意义(P<0.05)。qRT-PCR结果显示,在联合移植组中CD16、CD56、NKG2D mRNA的相对表达量较单独移植组明显增加,差异具有统计学意义(P<0.05)。结论NK细胞参与肝细胞性肝癌合并Eg感染裸鼠的移植瘤形成过程。
Objective To investigate the expression of nature killer(NK)cells in the transplanted tumor of hepatocellular carcinoma nude mice infected with Echinococcus granulosus(Eg).Methods Forty nude mice were randomly divided into control group and co-transplantation group.Mice in control group were subcutaneously transplanted with 5×10^(6) HepG2 cells,while mice in co-transplantation group were subcutaneously transplanted with 5×10^(6) HepG2 cells and 2000 Eg protoscolices(PSCs).Volume of subcutaneous lesions was measured every four days for five times from the 25 th day after transplantation and samples were obtained at 37 d after transplantation.The pathological changes were observed by hematoxylin and eosin(HE)staining.Expression of CD16,CD56 and NKG2D in the subcutaneous lesion was analyzed by immunohistochemical staining and real-time quantitative polymerase chain reaction(qRT-PCR)analysis.Results Compared with control group,the volume of subcutaneous lesions in co-transplantation group was significantly increased at different time(P<0.05).Heterogeneous cells in morphology and volume were found in control group,and the cytoplasm was alkalophilic.In addition,nucleoplasm ratio and mitotic activity were increased,and there was fatty degeneration,coagulative necrosis and vacuoles in some cells.Notably,those above pathological changes were more obvious and typical structures of Eg PSCs were also found in co-transplantation group.Immunohistochemical staining showed that CD16,CD56 and NKG2D positive cells were located in the cytoplasm of subcutaneous lesions and displayed as brown in colour.Quantitative data demonstrated that the expression of CD16,CD56 and NKG2D in co-transplantation group was significantly higher than that in control group(P<0.05).Moreover,qRT-PCR analysis also showed that the mRNA expression of CD16,CD56 and NKG2D in co-transplantation group was significantly up-regulated compared with control group(P<0.05).Conclusion NK cells are involved in the transplanted tumor formation process in the concurrent
