噬菌体抗体库分类与筛选方法的研究

摘要 噬菌体抗体库因具有构建过程简单及筛选方法高效、快速等优势,近年来受到广泛的关注。按照抗体库的来源可将噬菌体抗体库分为免疫噬菌体抗体库和非免疫噬菌体抗体库两大类。免疫噬菌体抗体库主要来源于能够产生免疫应答的动物体。非免疫噬菌体抗体库主要是通过半合成或全合成的方式获得。按照筛选抗原的纯度可将噬菌体抗体库的筛选方法分为纯化抗原筛选法和非纯化抗原筛选法两大类。纯化抗原筛选法主要是通过使用纯化过的抗原对抗体库进行筛选,具有效率高、特异性强等特点。非纯化抗原筛选法是对无法获得纯化抗原或抗原背景较复杂的抗体库进行筛选的方法,具有随意性强的特点。本研究主要是探讨噬菌体抗体库的分类与筛选方法,以便为噬菌体抗体库的构建提供参考。 Phage antibody library has attracted extensive attention in recent years due to its advantages of simple construction process and efficient and rapid screening method.According to the source of antibody library,phage antibody library can be divided into immune phage antibody library and non-immune phage antibody library.Immunophage antibody libraries are mainly derived from animals capable of producing immune responses.The non-immune phage antibody library is mainly obtained by semi-synthetic or total-synthetic methods.The screening methods of phage antibody library can be divided into purified antigen screening method and unpurified antigen screening method according to the purity of screened antigens.Purified antigen screening method mainly uses purified antigen to screen the antibody library,which has the characteristics of high efficiency and strong specificity.The unpurified antigen screening method is a method to screen the antibody library which cannot obtain purified antigen or whose antigen background is complex.This study mainly discusses the classification and screening methods of phage antibody library,so as to provide reference for the construction of phage antibody library.
