摘要
噬菌体展示技术(PhageDisplayTechniques,PDT)是一种用于筛选和改造功能性多肽的生物技术。该技术作为筛选与多种靶分子(如抗体、酶类、细胞表面受体等)具有特异性亲和力或活性的肽的一个有效方法,自问世以来,已取得了很大的发展,并被广泛地应用于基因治疗、基因疫苗研究、抗原表位研究、药物设计、研究细胞信号传导等领域[1]。但该技术在两方面仍需进一步完善:(1)寻求更为有效的表达载体;(2)进一步完善筛选方法。
Phage Display Techniques(PDT)is a biotechnique used for screening and reconstructing functingal polypeptide. It's a effective method of select the peptide which has high affinities to many given targets(such as antibody,Enzyme,surface receptor of cell). It has been made much progress and are widely used in many fields, such as genetic treatment, the study of genetic vaccine, epitope mapping, drug discovery after its appearance. But it has to get more perfact in two sides:( 1 )Seek more effective expression vector;(2)Get more perfect selection methods. Many selection methods are reviewed in this article.
