摘要
本研究旨在评估黄腐酚与异黄腐酚对变形链球菌致龋毒力因子的影响.首先通过生长曲线和流式细胞术测定了黄腐酚与异黄腐酚对浮游细菌生长的影响,此外还研究了这两种化合物对细菌产酸性和耐酸性的影响,最后使用结晶紫染色法和扫描电子显微镜(SEM)来定量和观察生物被膜的结构.结果表明,黄腐酚与异黄腐酚对变形链球菌浮游细胞的最低抑菌浓度(MIC)为32μg/mL和1024μg/mL,亚MIC浓度的两种化合物还显著降低了变形链球菌的产酸性和耐酸性,同时黄腐酚与异黄腐酚对变形链球菌生物被膜的形成也具有一定的抑制作用.
This study aims to evaluate the effects of xanthohumol and isoxanthohumol on the cariogenic virulence factor of Streptococcus mutans.The effects of xanthohumol and isoxanthohumol on the growth of the planktonic bacteria were measured by growth curve and flow cytometry.In addition,the effects of these two compounds on the acid production and acid tolerance of the bacteria were also studied.Finally,the structure of the biofilm was quantitated and observed by crystal violet staining and scanning electron microscopy(SEM).The minimum inhibitory concentrations(MIC)of xanthohumol and isoxanthohumol against Streptococcus mutans were 32μg/mL and 1024μg/mL.The sub-MIC concentration of these two compounds also significantly reduced the acid production and acid tolerance of Streptococcus mutans.Meanwhile,xanthohumol and isoxanthohumol also had certain inhibitory effects on the formation of the biofilm of Streptococcus mutans.
