不同溶剂对槟榔壳多酚组分及生物活性的影响 被引量:5

Analysis of the Polyphenol Components and Bioactivities of Areca Husk Extract by Different Solvents

摘要 旨在研究传统溶剂(水、甲醇、乙醇、丙酮和乙酸乙酯)和环保型低共熔溶剂(DESs)对槟榔壳提取物的多酚含量及组成、抗氧化活性、α-葡萄糖苷酶抑制活性的影响。结果表明,50%丙酮槟榔壳提取液中多酚含量最高为15.36 mg GAE/g DW,3种DESs槟榔壳提取液中多酚含量在11.78~12.46 mg GAE/g DW之间,乙酸乙酯提取液中多酚最低。采用HPLC-ESI-DAD对其酚类成分进行了定性定量,其中表没食子儿茶素、DL-儿茶素和异鼠李素-3-O-半乳糖苷为槟榔壳提取液中的主要成分,且甜菜碱:乳酸和L-脯氨酸:乙二醇提取液中DL-儿茶素含量分别高达13.81、11.33 mg/g DW。此外,3种DESs提取液对1,1-二苯基-2-苦基肼(DPPH)、2,2’-联氮-双(3-乙基苯并噻唑啉-6-磺酸)二铵盐(ABTS)自由基清除能力、铁还原能力(FRAP)和还原力(PR)活性均优于传统溶剂。不同溶剂提取物对α-葡萄糖苷酶的抑制活性不同,其中50%丙酮、甜菜碱:乳酸是抑制α-葡萄糖苷酶最有效的提取溶剂。主成分和皮尔逊相关系数分析结果表明,不同溶剂对槟榔壳提取物中多酚和生物活性有显著影响。通过验证不同溶剂槟榔壳提取物的体外抗氧化以及降糖能力,对槟榔壳活性功能的评估及开发应用提供理论依据和技术参考。 This present work aims to investigate the influences of extraction solvents such as traditional solvents (water,methanol,ethanol,acetone and ethyl acetate) and eco-friendly deep eutectic solvents (DESs) on the phenolic content and composition,antioxidant activity,inhibition of α-glucosidase of areca husk extracts.The results showed that the areca husk extracts extracted by 50% acetone exhibited the highest total phenolic content 15.36 mg GAE/g DW,and the three DESs extracts had higher total phenolic content from 11.78~12.46 mg GAE/g DW,while that extracted by ethyl acetate had the lowest total phenolic content.The phenolic compositions were identified and quantified by HPLC-ESI-DAD,and epigallocatechin,and DL-catechin and isorhamnetin-3-O-galactoside were the main components in the areca husk extract,meanwhile,the content of DL-catechin in betaine:lactic acid and L-proline:ethylene glycol extract were as high as 13.81 and 11.33 mg/g DW,respectively.In addition,the three DESs extracts had stronger antioxidant activity on 1,1-diphenyl-2-picrylhydrazine (DPPH),2,2’-azino-bis(3-ethylbenzo thiazoline-6-sulphonic acid) (ABTS),ferric reducing antioxidant power (FRAP) and reducing power (PR) than traditional solvents.Different solvent extracts had different inhibitory activities on α-glucosidase,among them,50% acetone/betaine:lactic acid were found to be the most effective extraction solvent to inhibit α-glucosidase (IC50=69.82,91.21 mg/mL respectively).The results of principal components analysis and Pearson correlation coefficient analysis showed that different solvents had significant effects on polyphenols and biological activity of areca husk extract.By verification of antioxidant and hypoglycemic capacity assays in vitro,the evaluation of the active function of areca husk will be provide the foundation for their development and application.
