摘要
目的建立一种以甲基紫作为显色剂的体外清除羟自由基的试验方法,并探讨作用机理。方法反应体系受酸度、显色剂、金属离子、H2O_(2)以及反应时间的影响,试验确定了最佳反应条件pH值为4.5,5.15×10^(-5)mol·L^(-1) MV溶液1.0mL,1.5×10^(-4)mol·L^(-1) FeSO4溶液1.0mL,0.3%H_(2)O_(2)溶液1.0mL,室温5min,在582nm处测定吸光度降低值,对中药提取物的抗氧化活性进行分析测定。结果试验了5种中药提取物,以当归对·OH的清除效果为最高,清除率达45.28%,枸杞清除率最低仅为12.94%,并基于各类中药成分对可能的机理进行了探讨。结论该方法简单、快速、方便操作,适用于中药清除·OH的定量检测及抗氧化活性性能评价。
OBJECTIVE A method for scavenging hydroxyl radicalsin vitro with methyl violet was established,and its mechanism was explored.METHODS The reaction system was affected by acidity,chromogenic agent,metal ions,hydrogen peroxide and reaction time.The optimum reaction conditions were determined as the pH value is 4.5,5.15×10^(-5)mol·L^(-1) MV 1.0mL,1.5×10^(-4)mol·L^(-1) FeSO_(4) 1.0mL,0.3% H_(2)O_(2)1.0mL at 5min.The spectrophotometric reduction value was determined at 582nm.The dose-effect relationship was analyzed and the antioxidant activity of the extracts of traditional Chinese medicine was evaluated.RESULTS Angelica sinensis had the highest scavenging effect on·OH,with a scavenging rate of 45.28%,Lyciumbarbarumwas a lowest of 12.94%.The possible mechanism was also discussed based on the ingredients of various traditional Chinese medicines.CONCLUSIONS The method is simple,rapid and easy to operate,whichis suitable for the quantitative detection of scavenging·OH and the evaluation of antioxidant activity of traditional Chinese medicine.
