摘要 目的:探究沙棘油对人胃癌细胞增殖的影响。方法:使用CCK-8实验筛选出沙棘油助溶剂DMSO(二甲基亚砜)的安全使用剂量,使用助溶剂DMSO溶解沙棘油后干预人胃癌BGC-823细胞,通过CCK-8实验检测细胞增殖变化水平。使用Western blot实验检测沙棘油干预人胃癌BGC-823细胞后,细胞凋亡基因P53、BCL-2、BAX蛋白表达水平。结果:细胞增殖试验显示,≤0.1%助溶剂DMSO细胞培养液对胃癌BGC-823细胞活性无影响,使用DMSO溶解沙棘油干预胃癌细胞,当沙棘油浓度为2、5、10、20μg·mL^(-1)均能够较好的抑制BGC-823细胞的增殖(P<0.05)。Western blot结果显示,当沙棘油浓度为5、10、20μg·mL^(-1)时,P53蛋白表达量均较对照组增加,差异有统计学意义(P<0.05);当沙棘油浓度为2、5、10、20μg·mL^(-1)时,BCL-2蛋白表达量均较对照组明显降低(P<0.05),BAX蛋白表达量均较对照组增加(P<0.05)。结论:沙棘油可通过P53信号通路抑制人胃癌细胞增殖达到抗肿瘤作用。 Objective:To explore the effects of seabuckthorn oil on the proliferation of human gastric cancer cells.Methods:CCK-8 experiment was used to screen out the safe dosage of seabuckthorn oil cosolvent DMSO(dimethyl sulfoxide),and cosolvent DMSO was used to dissolve sea buckthorn oil after intervening in human gastric cancer BGC-823 cells,then the level of cell proliferation changes was detected by CCK-8 experiment.Western blot was used to detect the expression levels of apoptosis genes P53,BCL-2,and BAX after sea buckthorn oil interfered with human gastric cancer BGC-823 cells.Results:The cell proliferation test showed that DMSO cell culture medium of less than or equal to 0.1%cosolvent had no effect on the cell viability of gastric cancer BGC-823 cells.DMSO was used to dissolve sea buckthorn oil to interfere with gastric cancer cells.When the sea buckthorn oil concentration was 2,5,10 and 20μg·mL^(-1),it could well inhibit the proliferation of BGC-823 cells(P<0.05).Western blot results showed that compared with the control group,when the seabuckthorn oil concentration was 5,10,20μg·mL^(-1),the P53 protein expression was significantly increased;compared with the control group when the seabuckthorn oil concentration was 2,5,10 and 20μg·mL^(-1),the expression of BCL-2 protein was significantly reduced,and the expression of BAX protein was significantly increased(P<0.05).Conclusion:Seabuckthorn oil can inhibit the proliferation of human gastric cancer cells through the P53 signal pathway,and achieve anti-tumor effects.
