转BpGLK1基因白桦叶色变异规律及生长特性分析 被引量:5

摘要 通过开展转BpGLK1基因白桦叶色变异规律研究,探讨BpGLK1干扰表达株系叶绿素含量的降低对生长的影响。以2年生3个BpGLK1过表达株系(OE1~3)、7个干扰表达株系(RE1~7)为试材,采用PCR及qRT-PCR技术对目标基因进行分子检测,同时测定其叶色参数和叶绿素相对含量的时序变化规律,分析转基因白桦的生长特性。结果表明:2年生的转BpGLK1基因白桦中,导入的目标基因仍整合于转基因株系的基因组中,且BpGLK1干扰表达株系中BpGLK1显著下调,BpGLK1基因的低表达对2年生苗高生长无影响,7个干扰表达株系中有6个株系在整个生长期叶绿素含量低于WT株系,叶色亮度及黄色程度高于WT株系,其中RE1、RE7株系上述性状均优于其他株系,是后续城市园林绿化应用推广的首选株系。 The leaf color variation of transgenic BpGLK1 birch was studied,and the effect of the decrease of chlorophyll content in BpGLK1 interference expression strain on growth was discussed.Three two-year-old BpGLK1 overexpression strains(OE1~3)and seven interference expression strains(RE1~7)were used as materials.PCR and qRT-PCR were used for molecular detection of the target gene.The temporal changes of leaf color parameters and chlorophyll relative content were determined,and growth characteristics of transgenic birch were analyzed.The results showed that the target gene was still integrated into the genome of the transgenic lines in the two-year-old transgenic BpGLK1 birch.The expression of BpGLK1 was significantly down-regulated in BpGLK1 interference expression strains.Low expression of BpGLK1 gene had no effect on the growth of 2-year-old plant height.The chlorophyll content of six of the seven interference expression strains was lower than that of WT during the whole growth period,and the brightness and yellow degree of leaves were higher than those of WT.Among them,RE1 and RE7 were superior to other strains in the above characters,which would be the first choice for urban landscaping.
