基于网络药理学探讨补中益气汤对新型冠状病毒肺炎恢复期的作用机制 被引量:6

Study on the Mechanism of Buzhong Yiqi Decoction(补中益气汤)on the Recovery Period of COVID-19 Based on Network Pharmacology

摘要 目的:基于网络药理学探讨补中益气汤治疗新型冠状病毒肺炎(COVID-19)恢复期的分子机制。方法:通过中药系统药理学数据库和分析平台(TCMSP)检索补中益气汤的成分及靶点,基于ADME参数筛选主要成分,使用Cytoscape3.6.0软件构建中药-成分-靶点网络,在OMIM、GeneCards数据库中检索COVID-19靶点,绘制韦恩图得到交集靶点,导入STRING数据库,构建交集靶点的蛋白质-蛋白质相互作用(PPI)网络,生成分值最佳的MCODE组件。使用Cytoscape3.6.0软件的BisoGenet插件绘制疾病及药物PPI网络,并用Merge功能取交集,根据拓扑参数筛选出核心节点。通过Metascape平台进行基因本体与京都基因和基因组百科全书富集分析,绘制气泡图和通路-靶点网络图对结果可视化处理。结果:检索出8味中药的135个主要成分、258个药物靶点、446个疾病靶点,中药-成分-靶点网络得到槲皮素、β-谷甾醇、木犀草素等10个关键化合物,PPI网络筛选出351个核心靶点,发现10个关键靶点:STAU1、FUS、TARDBP、CUL4B、EEF1A1、EIF4A3、TUBG1、RPS3、RPL6、RPS2,核心靶点主要富集在爱泼斯坦-巴尔病毒感染、甲型流感、麻疹、军团杆菌病、系统性红斑狼疮病等疾病通路,及核糖体、病毒致癌、蛋白酶体等信号通路中。结论:补中益气汤可通过多靶点、多通路消除炎症,减轻肺部损伤以及调节免疫等方面对COVID-19恢复期发挥潜在的治疗作用,本研究为新药的研发和COVID-19恢复期的治疗提供了新思路和理论基础。 Objective:To explore the molecular mechanism of Buzhong Yiqi Decoction(补中益气汤)in the treatment of novel coronavirus pneumonia(COVID-19)during the recovery period based on network pharmacology.Methods:The components and targets of Buzhong Yiqi Decoction were retrieved from the TCMSP database.The main components were screened based on ADME parameters,and Cytoscape 3.6.0 software was used to construct a Chinese medicine-component-target network.The COVID-19 target was retrieved in the OMIM and GeneCards databases.We drew the Venn diagram to get the intersection target,imported the STRING database,constructed the PPI network of the intersection target,and generated the MCODE component with the best score.We used the BisoGenet plug-in of Cytoscape 3.6.0 software to draw the PPI network of disease and medicine,and used the Merge function to take the intersection.The core nodes were screened out according to the topological parameters.The GO and KEGG enrichment analysis were performed on the Metascape platform,and the bubble chart and the pathway-target network chart were drawn to visualize the results.Results:Total 135 main components,258 medicinal targets,446 disease targets of 8 Chinese medicines were retrieved,and 10 key compounds including quercetin,β-sitosterol,and luteolin,etc.were obtained through the network of Chinese medicine-component-target network.PPI network screened 351 core targets and found 10 key targets:STAU1,FUS,TARDBP,CUL4B,EEF1A1,EIF4A3,TUBG1,RPS3,RPL6,RPS2.The core targets are mainly enriched in Epstein-Barr Virus infection,influenza A,measles,Legionnaires'disease,systemic lupus erythematosus and other disease pathways,and ribosomal,viral carcinogenesis,proteasome and other signaling pathways.Conclusions:Buzhong Yiqi Decoction can eliminate inflammation through multiple targets and multiple pathways,reduce lung damage and regulate immunity,and play a potential therapeutic role in the recovery period of COVID-19.This study provides new ideas and theoretical basis for the development of n
