摘要
目的:探讨热敏灸对新生小鼠缺氧缺血性脑损伤的抗凋亡作用及机制。方法:出生7 d的新生小鼠,随机分为假手术组、模型组和艾灸组,其中艾灸组根据艾灸前后尾温的变化,再分为非热敏灸组和热敏灸组。采用右侧颈总动脉结扎联合缺氧的方法制备缺氧缺血性脑损伤新生小鼠模型,HE染色观察脑组织形态结构,TUNEL染色检测脑组织细胞凋亡,免疫组织化学法检测caspase-9表达,放射免疫分析法测定脑组织caspase-3含量。结果:与假手术组相比,模型组小鼠脑梗死灶增大,脑组织排列疏松,脑组织凋亡细胞数和caspase-9表达显著增多(P<0.05),caspase-3含量增加;与模型组相比,非热敏灸组和热敏灸组小鼠脑梗死灶均缩小,脑组织排列较致密,脑组织凋亡细胞数和caspase-9表达均减少(P<0.05),caspase-3含量显著减少(P<0.05),尤以热敏灸组更为明显。结论:热敏灸可能通过降低脑组织caspase-9表达和减少caspase-3含量,减少细胞凋亡,从而达到减轻新生小鼠缺氧缺血性脑损伤。
Objective:To investigate the anti-apoptotic effect and mechanism of heat-sensitive moxibustion(HSM)in neonatal mice with hypoxic-ischemic(HI)brain injury.Methods:Neonatal mice(aged 7 days)were randomly divided into the sham group,the HI group and the moxibustion group;the moxibustion group was subdivided into non-heat-sensitive moxibustion(NHSM)group and heat-sensitive moxibustion group according to the difference of tail temperature of mice before and after the moxibustion.Neonatal hypoxic-ischemic encephalopathy(HIE)model was induced by ligation of the right common carotid artery combined with hypoxia in a closed transparent chamber.Themorphological structure of brain tissue was observed by HE staining,the apoptosis of brain tissue was detected with TUNEL assay,the expression of caspase-9 was detected by immunofluorescent staining,and the content of caspase-3 was determined by radioimmunoassay.Results:Compared to those in the sham group,the infarct areas increased,the brain tissues arranged loosely,the number of apoptotic cells increased and the expressions of caspase-9 and caspase-3 increased in the HI group(P<0.05);compared to those in the HI group,the infarct areas decreased,the brain tissues arranged tightly,the number of apoptotic cells decreased,the expressions of caspase-9 and caspase-3 decreased in both the NHSM group and the HSM group(P<0.05);of which the improvements were more significant in the HSM group.Conclusion:HSM can reduce neonatal hypoxic-ischemic brain injury,which may be related to reducing apoptosis through decreasing the expressions of caspase-9 and caspase-3 in brain tissues.
