针刺减少缺氧缺血性脑病新生小鼠脑组织炎性细胞浸润的研究 被引量:3

Study on acupuncture reducing inflammatory cell infiltration in brain tissue of neonatal mice with neonatal hypoxic-ischemic encephalopathy

摘要 目的:研究针刺减少缺氧缺血性脑病(HIE)新生小鼠脑组织炎性细胞浸润。方法:将出生7d的新生ICR小鼠随机分为假手术组、模型组和针刺组3组。采用颈总动脉结扎术联合缺氧制备缺氧缺血性脑损伤新生小鼠模型;TTC染色检测脑梗死面积;DAPI染色观察脑组织形态结构;HE染色观察脑组织炎细胞浸润;比色法测定髓过氧化物酶(MPO)活性;荧光免疫组织化学法检测脑组织CD11b表达。结果:与假手术组比较,模型组小鼠的脑梗死面积增大,患侧脑组织大量细胞坏死脱落,并伴有大量炎细胞浸润,MPO活性增高,CD11b阳性细胞数显著增多;与模型组比较,针刺组小鼠脑梗死面积缩小,患侧脑组织细胞排列较紧密、整齐,炎细胞较少,MPO活性有所降低,CD11b阳性细胞数明显减少。结论:针刺能够减轻新生小鼠缺氧缺血性脑损伤的作用,很可能与其减少脑组织炎性细胞(尤其是小胶质细胞)浸润、减轻炎症反应有关。 Objective:To observe the effect of acupuncture on inflammatory cell infiltration in brain tissues of neonatal hypoxic-ischemic encephalopathy(HIE)neonatal mice.Methods:Seven-day-old neonatal ICR mice were randomly divided into three groups,sham group,model group and acupuncture-treated group.Neonatal hypoxic-ischemia brain injury was induced by ligation of common carotid artery followed by hypoxia.TTC staining was used to examine infarct volume of brain,DAPI staining was used to observe morphology and structure,HE staining was used to detect inflammatory cells,then spectrophotometery was used to determine MPO activity and immunofluorescent staining was used to examine the CD11b-positive cells in the ipsilateral brain.Results:Mice in model group showed larger infarct area,more cells loss,more inflammatory cell infiltration,higher MPO activity,and more CD11b-positive cells in the ipsilateral brain than those in sham group,while mice in acupuncture-treated group showed smaller infarct area,less cell loss,less inflammatory cells,lower MPO activity,and less CD11b-positive cells in the ipsilateral brain than those in model group.Conclusion:Acupuncture may decrease microglia infiltration and alleviate inflammatory reaction,thereby reducing hypoxic-ischemic brain injury after HI procedure.
