摘要
目的探讨miR-30-5p和细胞因子信号传导抑制因子1(SOCS1)在食管癌组织中的表达及临床意义。方法选取2016年2月—2017年1月收治的食管癌85例,比较食管癌组织和癌旁组织中miR-30-5p及SOCS1 mRNA和蛋白表达,分析食管癌组织miR-30-5p和SOCS1 mRNA表达与食管癌患者临床病理特征关系,探讨食管癌组织中miR-30-5p和SOCS1 mRNA表达的相关性,预测食管癌组织中miR-30-5p和SOCS1 mRNA的结合位点,分析食管癌组织中miR-30-5p和SOCS1 mRNA不同表达与食管癌患者预后关系。结果食管癌组织中miR-30-5p相对表达量高于癌旁组织,SOCS1 mRNA相对表达量低于癌旁组织,SOCS1蛋白表达阳性率低于癌旁组织,差异有统计学意义(P<0.01)。食管癌患者不同病理分级、肿瘤分期和有无淋巴结转移食管癌组织miR-30-5p和SOCS1 mRNA表达差异有统计学意义(P<0.05或P<0.01)。食管癌组织中miR-30-5p和SOCS1 mRNA表达呈明显负相关(P<0.01)。SOCS1 mRNA序列3'非翻译区的285~292碱基位置处含有与miR-30-5p互补结合位点。3年总体生存率食管癌组织中miR-30-5p高表达患者低于低表达患者,SOCS1 mRNA高表达患者高于低表达患者,差异有统计学意义(P<0.01)。结论食管癌组织中miR-30-5p表达升高,而SOCS1 mRNA和蛋白表达降低,其与食管癌患者病理分级、肿瘤分期、有无淋巴结转移及预后有关,共同促进食管癌恶性进展。
Objective To study expressions of miR-30-5p and suppressor of cytokine signaling 1(SOCS1)in esophageal cancer and their clinical significances.Methods A total of 85 patients with esophageal cancer admitted between February 2016 and January 2017 were selected as research subjects.The mRNA and protein expressions of miR-30-5p and SOCS1 between esophageal cancer tissues and adjacent normal tissues were compared.Relationships between miR-30-5p and SOCS1 mRNA expressions with clinicopathological characteristics of esophageal cancer patients were analyzed,and the correlation between miR-30-5p with SOCS1 mRNA expressions was explored.The binding site of miR-30-5p and SOCS1 mRNA was predicted,and the correlations between different mRNA expressions of miR-30-5p and SOCS1 with the prognosis of esophageal cancer patients were analyzed.Results In esophageal cancer tissues,the relative expression of miR-30-5p was significantly higher than that in adjacent tissues,while relative mRNA expression of SOCS1 was significantly lower than that in adjacent tissues,and the positive rate of SOCS1 protein expression was significantly lower than that in adjacent tissues(P<0.01).There were statistically significant differences in mRNA expressions of miR-30-5p and SOCS1 in esophageal cancer tissues of patients with different pathological grades,tumor stages and presence or absence of lymph node metastasis(P<0.05 or P<0.01).The mRNA expressions of miR-30-5p and SOCS1 in esophageal cancer tissues were significantly negatively correlated(P<0.01).The SOCS1 mRNA sequence contained complementary binding sites with miR-30-5p at 285-292 bases in the 3'UTR region.The 3-year overall survival rate of esophageal cancer patients with high miR-30-5p expression was significantly lower than that in patients with low miR-30-5p expression,while the rate of esophageal cancer patients with high SOCS1 mRNA expression was significantly higher than that in patients with low SOCS1 mRNA expression(P<0.01).Conclusion The expression of miR-30-5p is increased,while th
