盐酸小檗碱对胃癌SGC-7901细胞增殖、凋亡、自噬及MAPK通路影响的研究 被引量:8

摘要 目的观察盐酸小檗碱对胃癌SGC-7901细胞增殖、凋亡、自噬及丝裂原活化蛋白激酶(MAPK)通路的影响,探讨盐酸小檗碱治疗胃癌的可能分子机制。方法通过CCK-8实验检测0.5,1,1.5,2,2.5,3,3.5 g/L的盐酸小檗碱培养24 h、48 h、72 h后胃癌SGC-7901细胞增殖情况,采用流式细胞技术检测0 g/L(空白组)、4 g/L和8 g/L的盐酸小檗碱培养48 h后胃癌SGC-7901细胞凋亡情况,采用Western blot实验检测0 g/L(空白组)、4 g/L和16 g/L的盐酸小檗碱培养12 h后胃癌SGC-7901细胞中Bax、Caspase-3、Bcl-2、LC3Ⅰ、LC3Ⅱ、Beclin 1、RAS、p38蛋白表达情况。结果从2 g/L盐酸小檗碱浓度开始,胃癌SGC-7901细胞存活率随着盐酸小檗碱浓度的增加而逐渐降低,差异均有统计学意义(P均<0.05);胃癌SGC-7901细胞凋亡率随着盐酸小檗碱浓度的增加而递增;与空白组比较,16 g/L盐酸小檗碱培养后胃癌SGC-7901细胞中Bax、Caspase-3、LC3Ⅱ、Beclin 1、RAS、p38表达量均显著增高,Bcl-2、LC3I蛋白表达量均显著减少,差异均有统计学意义(P均<0.05)。结论盐酸小檗碱可通过调控Bax/Bcl-2蛋白的表达抑制胃癌SGC-7901细胞增殖及促进其凋亡,可通过激活p38 MAPK信号通路诱导胃癌SGC-7901细胞发生自噬,从而达到对胃癌的干预和治疗作用。 Objective It is to observe the effects of berberine hydrochloride on the proliferation,apoptosis,autophagy and mitogen-activated protein kinase(MAPK)pathway of gastric cancer SGC-7901 cells,and to explore the possible molecular mechanism of berberine hydrochloride in the treatment of gastric cancer.Methods CCK-8 experiment was used to detect the proliferation of gastric cancer SGC-7901 cells after culturing with 0.5,1,1.5,2,2.5,3,3.5 g/L berberine hydrochloride for 24 h,48 h,and 72 h,using flow cytometry to detect the apoptosis of gastric cancer SGC-7901 cells after culture with 0 g/L(blank group),4 g/L,and 8 g/L berberine hydrochloride for 48 hours,Western blot was used to detect the expression of Bax,Caspase-3,Bcl-2,LC3Ⅰ,LC3Ⅱ,Beclin 1,RAS,and p38 protein in gastric cancer SGC-7901 cells after culturing with berberine hydrochloride at 0 g/L(blank group),4 g/L and 16 g/L for 12 hours.Results Starting from the concentration of 2 g/L berberine hydrochloride,the survival rate of gastric cancer SGC-7901 cells gradually decreased as the concentration of berberine hydrochloride increased,and the differences were statistically significant(all P<0.05);the cell apoptosis rate of gastric cancer SGC-7901 increased with the increase of berberine hydrochloride concentration;compared with the blank group,the expression levels of Bax,Caspase-3,LC3Ⅱ,Beclin 1,RAS,p38 in gastric cancer SGC-7901 cells after culture with 16 g/L berberine hydrochloride significantly increased,and the expression levels of Bcl-2 and LC3Ⅰprotein significantly reduced,and the differences were statistically significant(all P<0.05).Conclusion Berberine hydrochloride can inhibit the proliferation of gastric cancer SGC-7901 cells and promote their apoptosis by regulating the expression of Bax/Bcl-2 protein,and can induce autophagy in gastric cancer SGC-7901 cells by activating the p38 MAPK signaling pathway,so as to achieve the intervention and treatment of gastric cancer.
