摘要
目的分析2型糖尿病(T2DM)合并牙周感染患者白细胞介素-1β(IL-1β)、解偶联蛋白3(UCP3)基因多态性。方法选取三所医院T2DM伴牙周炎患者80例(研究组)、单纯T2DM患者75例(糖尿病组)、单纯牙周炎患者60例(牙周炎组),测定血清及龈沟液中IL-1β、UCP3水平,检测IL-1β-511、IL-1β+3953及UCP3-G304A、UCP3-55C/T基因位点单核苷酸多态性,分析IL-1β及UCP3基因多态性与T2DM合并牙周感染的相关性。结果研究组血清及龈沟液中IL-1β、UCP3表达水平高于糖尿病组、牙周炎组,牙周炎组血清及龈沟液中IL-1β、UCP3表达水平也较糖尿病组高(P<0.05);研究组IL-1β-511位点基因型AA及等位基因A频率高于糖尿病组、牙周炎组,研究组IL-1β+3953位点基因型ⅡⅡ及等位基因Ⅱ频率高于糖尿病组、牙周炎组(P<0.05),三组UCP3-G304A位点基因多态性比较,无统计学差异,研究组UCP3-55C/T位点基因型CC及等位基因C频率均较糖尿病组、牙周炎组低(P<0.05);IL-1β-511位点等位基因A、IL-1β+3953位点等位基因Ⅱ为T2DM患者发生牙周感染的危险因素,UCP3-55C/T位点等位基因C为保护因素(P<0.05)。结论IL-1β及UCP3基因多态性与T2DM并发牙周感染有密切联系,其中IL-1β+3953位点等位基因A、IL-1β+3953位点等位基因Ⅱ是危险因素,而UCP3-55C/T位点等位基因C为保护因素。
OBJECTIVE To analyze the gene polymorphisms of interleukin-1β(IL-1β)and uncoupling protein 3(UCP3)in the type 2 diabetes mellitus(T2 DM)patients complicated with periodontal infection.METHODS The patients were enrolled from three hospitals,totally 80 T2 DM patients complicated with periodontitis were assigned as the study group,75 patients with simple T2 DM were assigned as the diabetes group,and 60 patients with simple periodontitis were assigned as the periodontitis group.The levels of serum and gingival crevicular fluid IL-1βand UCP3 were determined,the single nucleotide polymorphisms of IL-1β-511,IL-1β+3953,UCP3-G304 A and UCP3-55 C/T gene loci were detected,and the correlation between the gene polymorphisms of IL-1βand UCP3 and the T2 DM complicated with periodontal infection was observed.RESULTS The expression levels of serum and gingival crevicular fluid IL-1βand UCP3 of the study group were significantly higher than those of the diabetes group and the periodontitis group,and the expression levels of serum and gingival crevicular fluid IL-1βand UCP3 of the periodontitis group were significantly higher than those of the diabetes group(P<0.05).The frequencies of genotype AA at IL-1β-511 locus and allele A of the study group were significantly higher than those of the diabetes group and the periodontitis group,and the frequencies of genotypeⅡⅡat IL-1β+3953 locus and alleleⅡof the study group were significantly higher than those of the diabetes group and the periodontitis group(P<0.05).There was no significant difference in the gene polymorphism at UCP3-G304 A locus among the three groups;the frequencies of genotype CC at UCP3-55 C/T locus and allele C of the study group were significantly lower than those of the diabetes group and the periodontitis group(P<0.05).The allele A at IL-1β-511 locus and alleleⅡat IL-1β+3953 locus were the risk factors for periodontal infection in the T2 DM patients,and the allele C at UCP3-55 C/T locus was the protective factor(P<0.05).CONCLUSION The gene polymo
