家蚕微孢子虫伴侣蛋白CCTθ的鉴定

摘要 家蚕微粒子病是一种严重危害蚕种生产的蚕病,被列为蚕桑生产唯一的检疫对象。分子伴侣协助底物蛋白折叠、装配和转运,但目前对家蚕微孢子虫分子伴侣CCT(chaperonin containing tailless complex polypeptide)的研究较少。本研究克隆了家蚕微孢子虫NbCCTθ基因,构建原核表达载体pET-28a-NbCCTθ表达重组蛋白,纯化后制备多克隆抗体。间接免疫荧光分析结果显示,NbCCTθ在家蚕微孢子虫的整个生命周期中均表达,但在不同发育阶段分布不同;在成熟孢子中主要分布于细胞质和质膜上,在裂殖增殖期主要分布于细胞质中,孢子形成期后期定位在细胞核中。研究结果为进一步探索NbCCTθ的具体功能奠定了基础,对家蚕微粒子病的防治也有一定的指导意义。 The pebrine is a silkworm disease that seriously endangers the production of silkworm eggs,and is listed as the only quarantine object for sericultural production.Molecular chaperones can assist in the folding,assembly and transport of substrate proteins,howerver,there are few studies on CCT(chaperonin containing tailless complex polypeptide)protein of Nosema bombycis.In this study,the NbCCTθgene of Nosema bombycis was cloned,and the prokaryotic expression vector pET-28 a-NbCCTθwas constructed to express the recombinant protein,which was purified to prepare the polyclonal antibody.The results of indirect immunofluorescence analysis showed that NbCCTθwas expressed throughout the life cycle of Nosema bombycis,but the distribution was different in different developmental stages.It mainly distributed on the cytoplasm and plasma membrane in mature spores,mainly distributed in the cytoplasm in the proliferation stage,and mainly distributed in the nucleus in the anaphase of the sporulation stage.The study lays a foundation for further exploration of the specific functions of NbCCTθand also has a certain guiding significance for the prevention and treatment of silkworm pebrine disease.
