摘要
目的研究半夏泻心汤及各配伍组对胃肠道运动相关受体及Cajal间质细胞(ICC)内Ca^(2+)浓度的影响,探究半夏泻心汤调节慢性胃炎胃肠道功能的作用机制。方法建立寒热错杂型慢性胃炎大鼠模型。大鼠随机分为正常组、模型组、全方组、苦降组、辛开组、甘补组以及阳性药组,灌胃给药7 d。对胃组织进行HE染色观察病理变化,RTqPCR技术和Western blot法分别测定5-HT4、AchE、DA2、c-kit的mRNA和蛋白表达,免疫荧光法检测蛋白共表达位置及表达量,荧光探针Fluo-4-AM标记Ca^(2+),检测各组Cajal间质细胞中Ca^(2+)浓度变化情况。结果各药物组能不同程度改善大鼠慢性胃炎症状,提高5-HT4、AchE和c-kit的mRNA和蛋白的表达,降低DA2的mRNA和蛋白的表达。全方组调节5-HT4、AchE和c-kit的mRNA以及5-HT4和DA2蛋白表达的效果更显著。5-HT4、DA2、AchE和c-kit出现共表达,共表达部位在细胞膜。各药物组能不同程度降低Cajal间质细胞内Ca^(2+)的荧光强度,以全方组效果更为显著。结论半夏泻心汤及各个配伍组可通过调节5-HT4、DA2、AchE和c-kit的表达,降低Cajal间质细胞内的Ca^(2+)浓度,抑制Cajal间质细胞钙超载,防止Cajal间质细胞兴奋节律失常,从而调节胃肠道运动,改善大鼠慢性胃炎的症状。
AIM To explore the mechanism of Banxia Xiexin Decoction and its different compatibility groups in regulating gastrointestinal function of rats with chronic gastritis by observing the gastrointestinal motility related receptors and Ca^(2+)concentration in interstitial cells of Caja(ICC).METHODS The rat models of chronic gastritis were established with exposure to cold and hot stimuli.Rats were randomly divided into normal group,model group,whole prescription group,bitter group,pungent group,sweet group and positive drug group for 7-day intragastric administration.The rats had their pathological changes of gastric tissue observed by HE staining;their mRNA and protein expressions of 5-HT4,AchE,DA2 and c-kit measured by RT-qPCR and Western blot respectively;their co-expression location of 5-HT4,AchE,DA2 and c-kit proteins observed and determined by immunofluorescence;their Ca^(2+)concentration of ICC in each group detected by labeling Ca^(2+)with fluorescent probe Fluo-4-AM.RESULTS Each intervened group shared improved symptoms of chronic gastritis,increased expressions of mRNA and proteins of 5-HT4,AchE and c-kit,and decreased mRNA and protein expression of DA2 at different extent,among which the whole prescription group took the lead.Co-expression of 5-HT4,DA2,AchE with c-kit respectively,and the co-expression site on the cell membrane were observed.Moreover,reduction at different degrees in the fluorescence intensity of Ca^(2+)in ICC among the whole prescription and each compatibility group,and the most significant effect in the whole prescription group were noticed as well.CONCLUSION Banxia Xiexin Decoction and its different compatibility groups can regulate gastrointestinal motility and improve the symptoms of chronic gastritis in rats by regulating the expressions of 5-HT4,AchE,DA2 and c-kit,reducing Ca^(2+)concentration,inhibiting the calcium overload and preventing the excitatory allorhythmia of ICC.
