摘要
目的动态观察血府逐瘀汤对心肌缺血大鼠血清中血管内皮生长因子(VEGF)、血管内皮生长因子受体-1(VEGFR-1)、血管性血友病因子(VWF)、内皮糖蛋白(CD105)蛋白表达影响。方法结扎冠状动脉左前降支建立心肌缺血大鼠模型。将大鼠随机分为7组:正常组(不做处理)、假手术组(只穿线不结扎)、模型组(造模后灌胃等量生理盐水)、他汀组(腹腔注射阿托伐他汀0.9mg·kg^(-1)·d^(-1))和低、中、高3个剂量实验组(分别灌胃3.51,7.02,14.04g·kg^(-1)·d^(-1)血府逐瘀汤药液)。于灌胃前、灌胃第3天、灌胃第7天分别采血,ELISA检测血清中VEGF、VEGFR-1、VWF、CD105表达水平。结果空白组VEGF、VEGFR-1、VWF、CD105含量一直无明显变化,处于基线水平,波动平稳;模型组含量整体呈上升趋势,他汀组及低、中、高3个剂量组含量也呈上升趋势,但他汀组及低、中、高3个剂量组的上升幅度比模型组更大。结论血府逐瘀汤能明显升高心肌缺血大鼠血清中VEGF、VEGFR-1、VWF、CD105表达水平,其中以高剂量组、灌胃第7天效果最佳。
Objective To observe the effect of Xuefu Zhuyu Decoction on vascular endothelial growth factor(VEGF),vascular endothelial growth factor receptor(VEGFR-1),von Willebrand factor(VWF)and endoglin antibody(CD105)in serum of rats with myocardial ischemia.Methods The left anterior descending coronary artery was ligated to establish a rat model of myocardial ischemia.The rats were randomly divided into 7 groups:normal group(no treatment),sham operation group(threading only without ligation),model group(gavage after modeling,etc.)Normal saline),statin group(intraperitoneal injection of atorvastatin 0.9mg·kg^(-1)·d^(-1))and low,medium and high dose experimental groups(different 3.51,7.02,14.04 g·kg^(-1)·d^(-1)blood palace sputum soup liquid).Blood was collected before gavage,on the 3rd day and 7d after gavage.ELISA detects the expression levels of VEGF,VEGFR-1,VWF,and CD105 in serum.Results The contents of VEGF,VEGFR-1,VWF,and CD105 in the blank group have not changed significantly,and are at the baseline level with stable fluctuations;the contents of the model group have an overall upward trend,and the contents of the statin group and the low,medium and high dose groups also show an upward trend.However,the statin group and the low,medium,and high dose groups have a greater increase than the model group.Conclusion Xuefu Zhuyu Decoction can significantly increase the expression levels of VEGF,VEGFR-1,VWF and CD105 in the serum of rats with myocardial ischemia.Among them,the high-dose group and intragastric administration on the 7th day have the best effect.
