摘要
目的:研究长链非编码RNA LINC01006对前列腺癌细胞增殖、迁移及侵袭的影响,分析其表达水平与术后病理及患者预后的相关性。方法:采用RT-qPCR检测LINC01006在前列腺癌细胞、正常细胞系中的表达;转染LINC01006的腺病毒过表达载体和siRNA,分别采用细胞增殖试剂盒、Transwell侵袭实验、流式细胞术等检测细胞活力、增殖、侵袭及凋亡能力的变化;通过生物信息学分析预测LINC01006的作用靶点,转染验证预测结果;分别在裸鼠皮下注射转染腺病毒空载体和LINC01006过表达载体的前列腺癌细胞系,建立小鼠移植瘤模型,测定移植瘤的体积及质量;收集前列腺癌患者一般资料、随访资料,检测LINC01006在前列腺癌病理组织细胞中表达。结果:LINC01006在前列腺癌细胞系中表达升高;LINC01006沉默抑制了细胞的增殖、迁移和侵袭能力,加速了细胞凋亡率;抑癌微小RNA miR-34a-5p被LINC01006海绵吸附;LINC01006在前列腺癌组织中的表达水平与患者的年龄和肿瘤大小无关,与患者病理Gleason评分、术前PSA值及远处或淋巴结转移情况有关。结论:LINC01006通过激活miR-34a-5p促进PCa进展,便于预后评判。
Objective:To study the effect of long non-coding RNA LINC01006 on the proliferation,migration and invasion of prostate cancer cells,and analyze the correlation between the expression level of LINC01006 and postoperative pathology and prognosis of prostate cancer.Methods:RT-qPCR was used to detect the expression of LINC01006 in prostate cancer cells and normal cell lines.The adenovirus overexpression vector and siRNA of LINC01006 were transfected into prostate cancer cells.Cell proliferation kits,Transwell invasion experiments,flow cytometry and other methods were used to detect changes in cell viability,proliferation,invasion and apoptosis.The target gene of LINC01006 was predicted through bioinformatics analysis,and the double luciferase reporter gene analysis was used to verify the prediction results.Prostate cancer cell lines transfected with adenovirus empty vector and LINC01006 overexpression vector were subcutaneously injected into nude mice to establish a mouse transplanted tumor model,and determine the size and quality of the transplanted tumor.The general data and follow-up data of prostate cancer patients were collected also,and LINC01006 was detected by RT-qPCR in prostate cancer cells.Results:LINC01006 was highly expressed in the PCa cell line.LINC01006 silence inhibited cell proliferation,migration and invasion ability,and accelerated the rate of cell apoptosis.In addition,the tumor suppressor gene miR-34a-5p in PCa is adsorbed by the LINC01006 sponge.The expression level of LINC01006 in prostate cancer was not related to age and tumor size(P>0.05),but related to pathological grade(P<0.01)and lymph node positive(P<0.01).Conclusion:LINC01006 promotes the progression of PCa by activating miR-34a-5p,which is of significance in the prognosis of prostate cancer.
