摘要
目的研究秀丽莓原花青素(proanthocyanidins,RPC)对糖尿病小鼠胰腺组织的保护作用及其机制。方法造模成功的C57BL/6雄性糖尿病小鼠,随机分成模型组(Mod)、双胍组(Met)、RPCL组和RPCH组,给药7周。每周检测体重和每2周检测FBG 1次,小鼠处死前1周检测OGTT;7周后眼球取血,检测血浆中血脂、SOD活力和MDA含量;保留红细胞检测GHb水平;摘取小鼠胰腺组织,进行HE染色,观察其病理改变;Western blot检测胰腺组织中p-AKT/AKT、Nrf2和HO-1蛋白的表达。结果RPC能明显降低小鼠FBG、GHb、OGTT、血脂和氧化应激水平,增加p-AKT/AKT、Nrf2和HO-1表达并改善胰腺组织病理学形态。结论RPC对糖尿病小鼠胰腺组织具有保护作用,其机制可能与激活PI3K/AKT/Nrf2信号通路有关。
Aim To study the protective effect of proanthocyanidins extracted from Rubus amabilis Focke(RPC)on the pancreatic tissues of diabetic mice and the underlying mechanism.Methods C57BL/6 male mice were given a high-sugar and high-fat diet combined with an intraperitoneal injection of STZ to establish the diabetic model.The mice whose FBG were higher than 16.7 mmol·L-1 were considered to be successfully modeled.The mice were randomly divided into model group,metformin group,RPCL group and RPCH group(administration for 7 weeks).The body weight and FBG were detected weekly and biweekly.OGTT was detected at the end of the 6th week.Seven weeks later,blood was taken from the eyeballs.The plasma was collected for detecting blood lipid,SOD activity and MDA content.The red blood cells were collected for detecting GHb.The pancreatic tissues were extracted and stained with HE.Western blot was employed for detecting the expression of p-AKT/AKT,Nrf2 and HO-1 in pancreatic tissues.Results RPC significantly reduced the FBG,GHb,OGTT,plasma lipids and oxidative stress levels,elevated the expression levels of p-AKT/AKT,Nrf2 and HO-1 protein in pancreatic tissues and ameliorated the pathological morphology of pancreatic tissues.Conclusion RPC reduces the damage of pancreatic tissues through activating the PI3K/AKT/Nrf2 signaling pathway.
