摘要
拟环纹豹蛛Pardosa pseudoannulata是稻田害虫重要的自然天敌,在绿色防控中有重要作用。本研究基于拟环纹豹蛛转录组数据,使用微卫星鉴定工具软件(MISA)挖掘和分析其简单序列重复(SSR)位点。结果表明:拟环纹豹蛛转录组Unigene序列中共检测到6474个SSR位点,平均长度为12.36 bp,分布于4411条Unigene上。单核苷酸重复类型为拟环纹豹蛛转录组SSR位点的主导基序类型,共5025个,占总SSR位点的77.62%;在拟环纹豹蛛转录组序列识别的6474个SSR中共发现了53种重复基元,从单核苷酸到五核苷酸重复基元分别有2种、6种、27种、15种和3种,其中(A/T)基元类型占明显优势,占总SSR的74.44%。拟环纹豹蛛转录组SSR位点丰富,有较高的多态性潜能,可作为开发SSR标记的有效来源,为拟环纹豹蛛的遗传多样性分析、功能基因的开发利用等方面的研究奠定基础。
The wolf spider Pardosa pseudoannulata is an important biological control agent of rice pests and plays an important role in green prevention and control.In this study,based on the transcriptome data of P.pseudoannulata,we used MISA(microsatellite identification tool)to analyze its SSR(simple sequence repeat)loci.The results showed that a total of 6474 SSR loci were detected in the Unigene sequence of the transcriptome data,with an average length of 12.36 bp,distributed on 4411 Unigenes.The Mono-nucleotide repeat type were the dominanttype,its number was 5025,accounting for 77.62%of the total SSR loci.The 6474 SSR lociwere identified 53 repetitive motifs,there were 2,6,27,15 and 3 repetitive motifs from Mono-nucleotide to Hexa-nucleotide.(A/T)was dominant repetitive motif,accounting for 74.44%of the total SSR.The SSR loci of transcriptome in P.pseudoannulata were rich and high polymorphism potential.Our results can be used as an effective source for the development of SSR marker and provide foundation for research of genetic diversity and development functional genes of P.pseudoannulata.
