微小RNA-326对乳腺癌细胞增殖和侵袭的影响

Effect of miR-326 on proliferation and invasion in breast cancer cells

摘要 目的探究Notch受体2(Notch receptor 2,Notch2)在乳腺癌细胞中的作用及机制。方法取MCF-7细胞系培养,分别转染pc-NC、pc-Notch2、NC mimic和miR-326 mimic。以细胞计数试剂盒(cell counting kit-8,CCK-8)检测细胞增殖情况,以Transwell实验检测细胞侵袭能力,以蛋白质印迹(Western blot)法检测Notch2蛋白表达。结果72 h时,pc-NC组和pc-Notch2组的细胞增殖水平分别为0.61±0.06和0.77±0.07;侵袭细胞数分别为(57.33±4.92)和(75.33±6.24)个,差异有统计学意义(P<0.05)。NC mimic组以及miR-326 mimic中Notch2蛋白表达量分别为1.01±0.10和0.69±0.06,差异有统计学意义(P<0.05)。结论Notch2可显著促进乳腺癌细胞的增殖和侵袭,miR-326可显著抑制Notch2的表达。 Objective To investigate the role and mechnaism of Notch receptor 2(Notch2)in breast cancer cells.Methods MCF-7 cell lines were cultured and transfected with pc-NC,pc-Notch2,NC mimic and miR-326 mimic.Cell counting kit-8(CCK-8)assay was performed to detect cell proliferation.Transwell assay was performed to detect cell invasion.Expression of Notch2 protein was detected by Western blot.Results At 72 h,the cell activity of pc-NC group and pc-Notch2 group were 0.61±0.06 and 0.77±0.07;the number of invasion cells in pc-NC group and pc-Notch2 group were 57.33±4.92 and 75.33±6.24,with significant difference(P<0.05).The Notch2 protein expression in NC mimic group and miR-326 mimic group were 1.01±0.10 and 0.69±0.06,with significant difference(P<0.05).Conclusion Notch2 can significantly promote the proliferation and invasion of breast cancer cells,and Notch2 expression was significantly suppressed by miR-326.
