摘要
目的比较自主建立的C57BL/6J-Tg(HRAS)转基因小鼠与BALB/c J小鼠的杂交1代小鼠CB6F1-Tg(HRAS)Nifdc(简称CB6F1-NIFDC)与日本转基因小鼠模型杂交1代小鼠CBy B6F1-Tg(HRAS)2Jic(简称CB6F1-CIEA)的脏器及血液生理指标异同,丰富自建模型的基础数据。方法选取8周龄的CB6F1-NIFDC及CB6F1-CIEA各10只,雌雄各半,活体称体质量,随后解剖并测定其主要脏器质量及血液生理指标。结果自主建立的CB6F1-NIFDC及日本引进的CB6F1-CIEA小鼠模型的部分脏器质量和血生理参数有差异性,脏器质量结果表明,雌性小鼠的心、肝、肺、左右肾等指标存在一定差异;雄性小鼠的肝、肺、左右肾、胸腺、左右睾丸指标质量有差异。22项血液生理指标方面,雌性小鼠在RBC、HGB等7项指标差异极显著(P<0.01);而雄性小鼠在RBC、HCT等5项指标差异极显著(P<0.01)。结论本文测定了两种模型脏器指标及生理指标,为自建模型在新药安全评价实验中的应用提供参考。
Objective To determine the difference of organs weight and blood physiological of CB6 F1-Tg(HRAS)Nifdc Mice and CByB6 F1-Tg(HRAS)2 Jic Mice.Method For this experiment,we selected 8-week-old CB6 F1-NIFDC and CB6 F1-CIEA mice(10 females and males)of each sex,weighed body mass and subsequently dissected and assayed their major organ masses and blood physiology.Result There are some differents between the CB6 F1-NIFDC and CB6 F1-CIEA models in some parts of the organs and blood physiological parameters.Female mice showed some differences in heart,liver,lung and kidney,male mice showed differences in liver,lung,kidney,thymus and left and right testicular index mass.Among female mice,there were significant differences in 7 indicators(RBC,HGB,etc.)in 22 blood physiological parameters(P<0.01);among male mice,there were significant differences in 5 indicators(RBC,HCT,etc.)in 22 blood physiological parameters(P<0.01).Conclusion In this paper,the organ indicators and physiological indexes of the two models were measured,which may provide a reference for the application of the self-built model in the safety evaluation experiments of new drugs.
