摘要 目的:基于活性氧(ROS)/硫氧还蛋白结合蛋白(TXNIP)通路探讨紫草素对布鲁氏菌感染小鼠的保护作用。方法:将BALB/C小鼠随机分为五组,即对照组、模型组、紫草素组(50 mg/kg)、N-乙酰半胱氨酸(NAC)组(ROS抑制剂NAC 20 mg/kg)以及紫草素+NAC组(紫草素50 mg/kg+ROS抑制剂NAC 20 mg/kg)。除对照组外,其余组腹腔注射布鲁氏菌液构建小鼠感染模型。相应药物处理后,测定五组小鼠肝脾指数,检测腹腔积液及脾脏中载菌量,以酶联免疫吸附法检测血清γ干扰素(IFN-γ)、白细胞介素6(IL-6)和肿瘤坏死因子α(TNF-α)水平,免疫球蛋白G(IgG)和免疫球蛋白M(IgM)水平,以ROS检测试剂盒检测血液中ROS水平,以蛋白免疫印迹法检测血液中TXNIP表达。结果:与对照组比较,模型组小鼠的肝脏指数、脾脏指数、腹腔积液载菌量及脾脏载菌量,血清IFN-γ、IL-6及TNF-α水平,IgG、IgM水平,血液中ROS水平和TXNIP蛋白表达水平显著升高,差异均有统计学意义(P<0.05)。与模型组比较,紫草素组、NAC组和紫草素+NAC组小鼠的肝脏指数、脾脏指数、腹腔积液载菌量及脾脏载菌量,血清IFN-γ、IL-6及TNF-α水平,IgG、IgM水平,血液中ROS水平及TXNIP蛋白表达水平显著降低,差异均有统计学意义(P<0.05)。与紫草素组及NAC组分别比较,紫草素+NAC组小鼠上述指标水平均显著降低,差异均有统计学意义(P<0.05)。结论:紫草素可降低布鲁氏菌感染小鼠的载菌量,减轻免疫炎症反应,对小鼠起到保护作用。 OBJECTIVE:To probe into the protective effect of shikonin on mice infected by brucella based on reactive oxygen species(ROS)/thioredoxin interacting protein(TXNIP).METHODS:BALB/C mice were randomly divided into five groups,including control group,model group,shikonin group(50 mg/kg),N-acetylcysteine(NAC)group(20 mg/kg of ROS inhibitor NAC)and paclitaxel+NAC group(50 mg/kg of paclitaxel,20 mg/kg of ROS inhibitor NAC).Except for the control group,the mouse infection model was constructed by intraperitoneal injection of Brucella liquid in the rest groups.After corresponding drug treatment,the liver and spleen indicators of mice in each group were measured,the bacterial load in peritoneal effusion and spleen were detected,levels of serum interferonγ(IFN-γ),interleukin 6(IL-6),tumor necrosis factorα(TNF-α),immunoglobulin G(IgG)and immunoglobulin M(IgM)were detected by enzyme-linked immunosorbent assay,the ROS level in blood was detected by ROS assay kit,and the expression of TXNIP in blood was detected by protein immunoblotting.RESULTS:Compared with the control group,the liver indicator,spleen indicator,bacterial load in peritoneal effusion and spleen,serum IFN-γ,serum IL-6,serum TNF-α,IgG and IgM levels,ROS level and expression level of TXNIP protein of mice in the model group were significantly higher,with statistically significant differences(P<0.05).Compared with the model group,the liver indicator,spleen indicator,bacterial load in peritoneal effusion and spleen,serum IFN-γ,serum IL-6,serum TNF-α,IgG and IgM levels,ROS level and expression level of TXNIP protein of mice in the shikonin group,NAC group and paclitaxel+NAC group were significantly lower,with statistically significant differences(P<0.05).Compared with shikonin group and NAC group,the above indicators in paclitaxel+NAC group were significantly lower,with statistically significant differences(P<0.05).CONCLUSIONS:Shikonin can reduce the bacterial load of Brucella in infected mice,alleviate immune inflammatory response,and play a protective role
