摘要
目的:研究卷曲乳酸杆菌对宫颈癌细胞增殖和凋亡的影响。方法:用卷曲乳酸杆菌与SiHa细胞共培养6 h、12 h、18 h及24 h,CCK-8法检测卷曲乳酸杆菌对SiHa细胞增殖的影响;流式细胞术检测卷曲乳酸杆菌对SiHa细胞凋亡的影响;酶联免疫吸附试验(ELISA)法检测细胞上清液中白细胞介素-2(IL-2)的含量;实时荧光定量PCR(qPCR)法检测增殖细胞核抗原(PCNA)mRNA相对表达量。结果:与卷曲乳酸杆菌共培养12 h开始,SiHa细胞增殖活性和PCNA mRNA相对表达量均显著降低,共培养6 h开始SiHa细胞凋亡率显著升高,共培养6 h和12 h细胞上清液中IL-2含量显著升高,而共培养24 h显著下降(均P<0.01)。结论:卷曲乳酸杆菌可显著抑制SiHa细胞的增殖并促进其凋亡。
Objective:To study the effect of Lactobacillus crimpus on the proliferation and apoptosis of cervical cancer SiHa cells.Methods:SiHa cells were co-cultured with Lactobacillus crimpus for 6 h,12 h,18 h and 24 h.CCK-8 method was used to detect the proliferation of SiHa cells.Flow cytometry was used to detect the apoptosis rate of SiHa cells.The content of IL-2 in the supernatant was detected by enzyme-linked immunosorbent assay(ELISA).The mRNA expression of proliferating cell nuclear antigen(PCNA)was detected by qPCR.Results:After co-culture with Lactobacillus crimpusfor 12 h,the proliferation of SiHA cells and the expression of PCNA mRNA were significantly decreased,while the apoptosis rate of SiHA cells was significantly increased after co-culture for 6 h,the content of IL-2 in the supernatant was significantly increased after co-culture for 6 h and 12 h,but it was significantly decreased after co-culture for 24 h(P<0.01).Conclusion:Lactobacillus crimpus can significantly inhibit the proliferation and promote the apoptosis of SiHa cells.
