摘要
目的:建立肉苁蓉配方颗粒和破壁饮片的UPLC指纹图谱及多成分含量测定方法,为其质量评价提供依据。方法:采用ACQUITY UPLC BEH C18(100 mm×2.1 mm,1.7μm)色谱柱,以乙腈-0.1%磷酸溶液为流动相,梯度洗脱,流速0.3 mL·min^(-1),检测波长235 nm,柱温35℃,进样量1μL。结果:建立了肉苁蓉配方颗粒和破壁饮片的UPLC指纹图谱,确定了10个共有峰,指认了京尼平苷酸、咖啡酸、松果菊苷、毛蕊花糖苷、管花苷A和异毛蕊花糖苷6种成分,并对这6种成分进行了含量测定,各成分分离良好,且有较宽的线性范围和良好的线性关系,稳定性、重复性及回收率均符合要求。采用聚类分析和主成分分析方法可以对肉苁蓉配方颗粒和破壁饮片进行区分和鉴别。结论:UPLC指纹图谱结合多成分含量测定方法简便、准确,重复性良好,可快速鉴别肉苁蓉配方颗粒和破壁饮片并评价其内在质量,为完善肉苁蓉配方颗粒和破壁饮片的质量控制提供方法参考。
Objective:To establish the UPLC fingerprints and multi-components content determination method of Cistanches herba formula granules and ultrafine granular power,and to provide a basis for their quality evaluation.Methods:The separation was achieved by using a ACQUITY UPLC BEH C18(100 mm×2.1 mm,1.7μm)column via gradient elution with the mobile phase consisting acetonitrile and 0.1%phosphoric acid water.The flow rate was 0.3 mL·min^(-1) and the detection wavelength was set at 235 nm;the column temperature was 35℃and the injection volume was 1μL.Results:The UPLC fingerprints of Cistanches herba formula granules and ultrafine granular power were established,in which 10 common peaks were determined,and 6 components of geniposidic acid,caffeic acid,echinacoside,acteoside,tubuloside A,and isoacteoside were identified.The contents of these 6 components were determined.Using the developed method,the components were well separated,and had wide linear ranges with good linear relationships.The stability,repeatability and recovery rate met the requirements.Using cluster analysis and principal component analysis methods,Cistanches herba formula granules and ultrafine granular power can be distinguighed and identified.Conclusion:The UPLC fingerprints combined with multicomponents content determination method are simple,accurate,and reproducible.It can quickly identify the formula granules and ultrafine granular power of Cistanches herba and evaluate their internal quality,providing a method reference for improving the quality control of formula granules and ultrafine granular power of Cistanches herba.
