当归内酯对鸡新城疫疫苗免疫效价、免疫细胞及相关因子的影响 被引量:6

Effects ofα-angelica lactone on immune titer,immune cells and related factors of Newcastle disease vaccine

摘要 第1批240只7日龄青脚麻鸡随机分为6组,Ⅰ组为空白对照,Ⅱ组为疫苗对照,Ⅱ、Ⅵ组在14,28日龄接种新城疫弱毒疫苗,Ⅲ~Ⅵ组于每次免疫前连续皮下注射当归内酯3d,剂量分别为10,20,40,80mg/kg,Ⅰ、Ⅱ组注射等量生理盐水。各组分别于免疫前和免疫后7,14,21,28,35d采集心血,检测血清新城疫抗体效价,并测定免疫器官指数。第2批120只7日龄青脚麻鸡随机分为4组,Ⅶ组为空白对照,Ⅷ组为疫苗对照,Ⅷ~Ⅹ组于14,28日龄接种新城疫弱毒疫苗,Ⅸ组在免疫前连续3d皮下注射当归内酯20mg/kg,Ⅹ组在免疫前连续5d注射当归内酯20mg/kg,Ⅶ、Ⅷ组注射等量生理盐水。各组于免疫前和免疫后7,14,21,28,35d采集心血,测定血液中T淋巴细胞亚群数量,并进行脾脏淋巴细胞增殖试验及细胞因子测定。结果显示,疫苗免疫后7,21d,Ⅳ组新城疫抗体效价显著高于Ⅱ组(P<0.05),免疫后7d法氏囊指数Ⅳ组显著高于Ⅱ组(P<0.05);注射20mg/kg当归内酯可提高鸡新城疫免疫抗体效价和法氏囊指数。Ⅹ组T、B淋巴细胞增殖显著高于Ⅶ、Ⅷ组(P<0.05),Ⅸ、Ⅹ组CD4^(+)及CD4^(+)/CD8^(+)比值显著高于Ⅶ~Ⅷ组(P<0.05),且Ⅹ组显著高于Ⅸ组(P<0.05);免疫后21,28d,Ⅸ组IL-2mRNA表达量显著高于Ⅷ~Ⅹ组(P<0.01);结果表明,20mg/kg当归内酯连续注射5d,可促进鸡T、B淋巴细胞增殖,提高CD4^(+)及CD4^(+)/CD8^(+)的比值,上调IL-2mRNA表达量。 The first batch of 2407-day-old green-footed chickens were randomly divided into 6 groups,groupⅠwas the blank control group,groupⅡwas the vaccine control group,and groupsⅡ-Ⅵwere vaccinated with attenuated Newcastle disease vaccine at the age of 14,28d.Ⅵgroup was subcutaneously injected withα-angelica lactone continuously for 3dbefore each immunization,and the doses were 10,20,40,80mg/kg,groupⅠand groupⅡwere injected with the same amount of normal saline.Heart blood was collected before immunization and 7,14,21,28,35dafter immunization,and the serum antibody titer of Newcastle disease and the immune organ index were determined.The second batch of 1207-day-old green-footed chickens were randomly divided into 4 groups,groupⅦwas the blank control group,groupⅧwas the vaccine control group,groupⅧand groupⅩwere vaccinated with attenuated Newcastle disease vaccine at the age of 14,28d,groupⅨwere subcutaneously injected with 20mg/kg ofα-angelica lactone for 3consecutive d before immunization,groupⅩwas injected with 20mg/kg ofα-angelica lactone for 5consecutive d before immunization,and groupsⅦandⅧwere injected with the same amount of normal saline.Heart blood was collected from each group before and 7,14,21,28,35dafter immunization to determine the number of T lymphocyte subsets in the blood,and splenic lymphocyte proliferation test was performed and cytokines were detected.The results showed that 7and 21after vaccination,the titers of Newcastle disease antibody in groupⅣwere significantly higher than those in groupⅡ(P<0.05);the bursal index of groupⅣwas significantly higher than that in groupⅡ(P<0.05)7dafter vaccination;20mg/kg ofα-angelica lactone injection could increase the titer of Newcastle disease immune antibodies and the bursa index of chickens;the proliferation of T and B lymphocytes in groupⅩwas significantly higher than that in groupsⅦandⅧ(P<0.05);the ratio of CD4^(+)and CD4^(+)/CD8^(+)in groupsⅨandⅩwas significantly higher than that in groupsⅦandⅧ(P<0.05
