两地牛蒡根总黄酮的体外和体内抗氧化活性研究 被引量:5

摘要 以江苏徐州A和山东苍山B两地的牛蒡根为原料,采用超声波辅助法提取牛蒡根中的总黄酮,并用大孔树脂联合聚酰胺法对其进行纯化后,从体外和体内2个方面研究牛蒡根总黄酮的抗氧化活性。结果表明,两地牛蒡根总黄酮均有很强的总抗氧化能力和铁离子还原力,其清除1,1-二苯基-2-三硝基苯肼(1,1-Diphenyl-2-picrylhydrazyl radical,DPPH)自由基、羟自由基、超氧阴离子自由基的半数抑制质量浓度IC_(50)分别为A:0.542±0.049 mg/mL,B:0.642±0.058 mg/mL;A:0.761±0.068 mg/mL,B:1.04±0.098 mg/mL;A:0.793±0.084 mg/mL,B:1.06±0.12 mg/mL。金属离子螯合能力IC_(50)分别为A:27.48±0.24μg/mL,B:29.40±0.32μg/mL,其能显著提升小鼠血清和肝脏的总抗氧化能力(Total antioxidization capacity,T-AOC)、过氧化氢酶(Catalase,CAT)活性、超氧化物歧化酶(Superoxide dismutase,SOD)活性,同时能显著降低小鼠血清和肝脏的丙二醛(malondialdehyde,MDA)含量,两地的牛蒡根总黄酮体内抗氧化活性没有显著差异。由此说明,两地的牛蒡根总黄酮均有较强的体外抗氧化能力和体内抗氧化能力,且没有显著差异。 The total flavonoids of burdock root from Xuzhou A and Cangshan B were extracted by the method of ultrasonic-assisted extraction and purified by macroporous resin and polyamide method,the antioxidant activity of total flavoniods of burdock root was studied in vitro and in vivo.The results showed that total flavonoids of burdock root showed a strong total antioxidant capacity and strong ferric reducing ability,DPPH radical scavenging activity(IC_(50) A:0.542±0.049 mg/mL,B:0.642±0.058 mg/mL),hydroxy free radicals scavenging activity(IC_(50) A:0.761±0.068 mg/mL,B:1.04±0.098 mg/mL),Superoxide anion radical scavenging activity(IC_(50) A:0.793±0.084 mg/mL,B:1.06±0.12 mg/mL)and ferrous ions chelating activity(IC_(50) A:27.48±0.24μg/mL,B:29.40±0.32μg/mL),the activity of T-AOC,CAT and SOD in serum and liver of mice was significantly increased,and the content of MDA in serum and liver of mice was significantly decreased by eating total flavonoids of burdock root from different places.That is to say,total flavonoids of burdock root have strong antioxidant activity in vivo and in vitro,and there was no significant difference in the antioxidant activity of total flavonoids in burdock root between the two regions.
