菜薹花枝离体培养留种技术研究

摘要 为了保证秋冬季低温寡照等不良条件下菜薹种子能正常成熟,从而提高菜薹的育种效率,研究剪取菜薹花枝扦插于特定营养液中,从营养液成分、培养条件等方面入手,探讨最适宜的花枝离体培养条件。结果表明:采用10%多抗霉素1 000倍液+0.1%农用链霉素+2%白糖+ 6-BA 2 mg/L的培养液,将扦插花枝置于25-28℃、日照10 h/d的光照培养箱中培养,对菜薹花枝生长最为有利,花枝不易滋生霉菌,可以正常吸收营养以供种子膨大发育成熟,成熟后种子饱满,发芽率可达98.2%。该花枝离体培养留种技术操作简便,有利于菜薹科研工作的顺利开展。 In order to ensure the normal maturity of Brassica campestris L.ssp.chinensis var.perperea seeds under adverse conditions such as low temperature and low illumination in autumn and winter,so as to improve the breeding efficiency of Brassica campestris L.ssp.chinensis var.perperea,its flower branches were cut and put into specific nutrient solution to explore the optimal in vitro culture conditions from the aspects of nutrient solution composition and culture conditions.The results showed that using 10%polyoxin 1000 times liquid+0.1%streptomycin+2%sugar+6-BA 2 mg/L as culture medium,the cuttings of flower branches were placed into the light incubator,maintaining sunshine 10 hours/day at 25-28°C,which was the most favorable for the growth of flower branches,so the flower branches were not easy to breed molds and could absorb nutrients normally for seed expansion and maturation.The seeds were plump after ripening and the germination rate was 98.2%.The technique of flower branch in vitro culture for seed reservation is simple and convenient,which is conducive to the smooth development of scientific research work of Brassica campestris L.ssp.chinensis var.perperea.
