摘要
目的建立银兰含漱方UPLC指纹图谱及其中6种成分的质量浓度测定方法,为银兰含漱方的质量控制提供依据。方法采用Phenomenex Luna Omega PS C_(18)(2.1 mm×150 mm,1.6μm)色谱柱;以乙腈-0.1%甲酸(含0.05%磷酸)为流动相,梯度洗脱;流速为0.40 mL/min;柱温为30℃;检测波长为245 nm;建立15批银兰含漱方的指纹图谱,通过指纹图谱的相似度分析结合主成分分析(PCA)和偏最小二乘法-判别分析(PLS-DA)进行评价,同时测定甘草苷、黄芩苷、汉黄芩苷、黄芩素、甘草酸、汉黄芩素6种成分的质量浓度。结果建立了银兰含漱方UPLC指纹图谱,标定了13个共有峰,各批次指纹图谱相似度均达0.9以上,PCA分析和PLS-DA分析显示各批样品无明显聚类趋势,表明银兰含漱方的批次一致性较高;甘草苷、黄芩苷、汉黄芩苷、黄芩素、甘草酸、汉黄芩素分别在29.20~292.00μg/mL、247.14~2471.40μg/mL、17.96~107.76μg/mL、18.16~181.60μg/mL、23.78~237.80μg/mL、12.47~124.70μg/mL范围内呈线性关系,R值均大于0.999,平均回收率分别为97.91%、100.85%、97.99%、100.01%、98.91%、98.85%。结论所建立的UPLC指纹图谱结合多成分定量分析的方法稳定、可靠,重复性好,为银兰含漱方的质量控制提供了一定的科学依据。
Objective To establish the UPLC fingerprint of Yinlan Hanshufang and simultaneous determination method of 6 components,so as to provide a reference for the quality control of Yinlan Hanshufang.Methods The column was Phenomenex Luna Omega PS column C_(18)(2.1 mm×150 mm,1.6μm)with a mobile phase of acetonitrile-0.1%formic acid(0.05%phosphoric acid)in gradient elution.The flow rate was 0.40 mL/min,the column temperature was 30°C,and the detection wavelength was set at 245 nm.The quality of 15 batches of Yinlan Hanshufang was evaluated by similarity analysis,PCA and PLS-DA of fingerprints,and the content of 6 components,including glycyrrhizin,baicalin,wogonoside,baicalein,glycyrrhizic acid and wogonin was determined simultaneously.Results The UPLC fingerprint of Yinlan Hanshufang was established,and thirteen peaks were selected as the character fingerprint peaks.The similarity of each batch of fingerprint was more than 0.9.PCA analysis and PLS-DA analysis showed that there was no obvious clustering trend in each batch of samples,indicating that the batch consistency of Yinlan Hanshufang was high.Good linear relationships were found in the range of 29.20-292.00μg/mL for glycyrrhizin,247.14-2471.40μg/mL for baicalin,17.96-107.76μg/mL for wogonoside,18.16-181.60μg/mL for baicalein,23.78-237.80μg/mL for glycyrrhizic acid,12.47-124.70μg/mL for wogonin,R values were all greater than 0.999,and the average recoveries were 97.91%,100.85%,97.99%,100.01%,98.91%,98.85%,respectively.Conclusion The established UPLC fingerprint and quantitative analysis method of Yinlan Hanshufang are stable,reliable and reproducible,which provides a scientific basis for the quality control of Yinlan Hanshufang.
