摘要
目的分析miR-21-5p在肾透明细胞癌组织和癌旁正常肾组织中的表达情况及其临床意义。方法收集27例肾透明细胞癌患者临床相关资料及其肾透明细胞癌和癌旁正常肾组织27对。采用RT-PCR法检测miR-21-5p在肾透明细胞癌和癌旁正常肾组织中的表达水平,进一步分析其表达水平与临床及病理特征的关系。应用生物信息学分析方法探讨miR-21-5p可能的作用机制。结果肾透明细胞癌组织中miR-21-5p的表达量高于癌旁正常肾组织,分别为(2.118±1.402)和(1.246±0.711),(P=0.014)。肾透明细胞癌组织中miR-21-5p的表达水平与患者性别、年龄、吸烟史、体重指数、TNM分期及病理核分级无明显关系(P>0.05);生存分析提示肾透明细胞癌组织中miR-21-5p上调表达和下调表达的患者发生疾病进展的比率无统计学差异(P>0.05)。生物信息学分析提示ADNP(activity dependent neuroprotector homeobox)等85种基因是miR-21-5p的靶基因,应用GO数据库和KEGG数据库发现muscle cell fate determination等63条可能的作用通路。结论 miR-21-5p在肾透明细胞癌患者癌组织中呈现高表达;生物信息学分析可为今后的实验研究提供线索和方向。
Objective To analyze the expression of miR-21-5p in renal clear cell carcinoma and adjacent normal renal tissue and its clinical significance. Methods The clinical data of 27 cases with renal clear cell carcinoma were collected. 27 pairs of renal clear cell carcinoma and adjacent normal renal tissue were collected too. The expression level of miR-21-5p in renal clear cell carcinoma tissues and adjacent normal renal tissues was detected by RT-PCR, and the relationship between the expression level and clinical and pathological characteristics was further analyzed. Bioinformatics analysis was used to investigate the possible mechanism of miR-21-5p. Results The expression level of miR-21-5p in renal clear cell carcinoma tissues was higher than that in adjacent normal renal tissues(2.118±1.402 and 1.246 ±0.711, respectively)(P = 0.014). The expression level of miR-21-5p in renal clear cell carcinoma tissues was not significantly correlated with gender, age, smoking history, body mass index, TNM stage and pathological nuclear grade(P> 0.05). Survival analysis indicated that there was no statistical difference in the rate of disease progression between the up-regulated and down-regulated miR-21-5p in renal clear cell carcinoma(P > 0.05). Bioinformatics analysis suggested that 85 kinds of genes such as ADNP(Activity dependent Neuroprotector homeobox)were the target genes of miR-21-5p. 63 kinds of possible pathways such as Muscle Cell Fate Determination pathway were found by GO database and KEGG database. Conclusion MiR-21-5p is highly expressed in the carcinoma tissues of patients with renal clear cell carcinoma. Bioinformatics analysis can provide clues and directions for future experimental research.
