摘要
目的探讨姜黄素在乙型病毒性肝炎中发挥抗纤维化作用的相关机制。方法制备SD大鼠乙型病毒性肝炎模型,将SD大鼠随机分为假手术组(Sham组)、模型组(HBV组)和姜黄素治疗组(Curcumin组);HE染色法检测各组大鼠肝脏的病理损伤情况;ELISA法检测血清中丙氨酸转氨酶(Alanine aminotransferase,ALT)、天冬氨酸转氨酶(Aspartateaminotransferase,AST)、透明质酸(Hyaluronic acid,HA)、层粘连蛋白(Laminin,LN)、肿瘤坏死因子(Tumornecrosisfactor,TNF-α)、白介素-6(Interleukin-6,IL-6)的水平变化;实时荧光定量PCR法对肝脏中HBV-DNA的表达水平进行检测;Western blot法检测肝脏中的过氧化物酶体增殖物激活受体γ(Peroxisome proliferatorsactivated receptorγ,PPAR-γ)、c-Ski(cellular Sloan-Kettering institute)、转化生长因子-β(Transforming growth factor-β,TGF-β)、Smad2、p-Smad2、Smad3和p-Smad3的表达状况。结果姜黄素可以显著改善肝组织损伤,降低肝功能损伤的相关指标和炎症因子的水平,抑制肝脏组织中HBV-DNA的表达,促进PPARγ、c-Ski的表达,抑制TGF-β、Smad2、pSmad2、Smad3、p-Smad3的表达。结论姜黄素在乙型病毒性肝炎中具有显著的抗纤维化的作用,其机制可能是通过激活PPAR-γ、c-Ski的表达,抑制TGF-β/Smads信号通路的活化来实现的。
Objective To explore the anti-fibrosis mechanism of Curcumin in hepatitis B. infection.Methods SD rats were randomly divided into Sham group, HBV group and Curcumin group. HE staining was used to detect the pathological damage of liver. The levels of ALT, AST, HA, LN, TNF-α and IL-6 were detected by ELISA. The expression of HBV-DNA in liver was detected by Real-time PCR;the expression of PPAR-γ, c-ski, TGF-β, Smad2, pSmad2, Smad3 and p-smad3 in liver was detected by Western blot. Results Curcumin can significantly improve liver injury, reduce the levels of related indicators and inflammatory molecules, inhibit the expression of HBV-DNA in liver tissue, promote the expression of PPAR-γ and c-Ski, and inhibit the expression of TGF-β, Smad2, p-Smad2, Smad3 and pSmad3. Conclusion Curcumin has a significant anti-fibrosis function in hepatitis B infection, which may be achieved by activating the expression of PPAR-γ and c-Ski, and inhibiting the expression of TGF-β/Smads signaling pathway.
