摘要
目的:建立川贝母生物碱贝母素甲的柱前紫外衍生—高效液相色谱法的测定方法。方法:以对甲苯磺酰氯为衍生试剂,在60℃的水浴条件下对贝母素甲水浴加热回流,采用HPLC-UV法测定川贝母中贝母素甲的含量。结果:在流动相为1 ml/min的甲醇—水(60∶40,v/v),柱温为35℃,检测波长为240 nm。贝母素甲在0.05~2.50μg/ml范围内线性相关系数为0.9993,方法检出限为0.008μg/ml和定量限为0.017 mg/ml,方法加标回收率为95.67%~100%,方法精密度(n=6)为2.4%。结论:本方法快速简便、灵敏度高、准确性好,适用于川贝母生物碱贝母素甲的含量测定。
Objective:To establish a method for the determination of Fritillaria thunbergii in Fritillaria thunbergii.Methods Using p-toluenesulfonyl chloride as derivative reagent,After 60℃of water bath,HPLC-UV method was used to determine the content of Fritillaria thunbergii.Results Methanol was 1 ml/min in mobile phase:water(60∶40,v/v),At 35℃,Detection wavelength 240 nm.The linear correlation coefficient of Fritillin A was 0.9993 in the range of 0.05~2.50μg/ml,The detection limit was 0.008μg/ml and the quantitative limit mg/ml,0.017 Method:The recovery rate was 95.67%~100%,method precision(n=6)was 2.4%.Conclusion:This method is fast,simple,sensitive and accurate,It is suitable for the determination of alkaloids in Fritillaria thunbergii.
