摘要
目的观察β-榄香烯对结肠癌SW480细胞迁移及侵袭能力的影响,以及Afadin蛋白磷酸化水平在其中发挥的作用。方法2019年10月—2020年1月于河北中医学院实验中心进行实验。将SW480细胞分为对照组、IGF-1组、LY294002组、榄香烯组、榄香烯+IGF-1组及榄香烯+LY294002组,分别接受β-榄香烯、IGF-1及LY294002作相应处理,应用Western-blot法检测各组细胞内Akt、磷酸化Akt、Afadin及磷酸化Afadin蛋白表达水平,应用划痕实验检测各组细胞迁移能力,应用Transwell侵袭实验检测各组细胞侵袭能力。结果与对照组比较,IGF-1组磷酸化Akt及磷酸化Afadin蛋白表达水平升高,LY294002组、榄香烯组上述蛋白表达水平降低;与榄香烯组比较,榄香烯+IGF-1组磷酸化Akt及磷酸化Afadin蛋白表达水平升高,榄香烯+LY294002组上述蛋白表达水平降低,差异均有统计学意义(P<0.05);各组结肠癌细胞Akt、Afadin蛋白表达水平比较,差异均无统计学意义(P>0.05)。与对照组比较,IGF-1组细胞迁移率、穿透基底膜的细胞数量升高,LY294002组、榄香烯组细胞迁移率、穿透基底膜的细胞数量降低;与榄香烯组比较,榄香烯+IGF-1组细胞迁移率、穿透基底膜的细胞数量升高,榄香烯+LY294002组细胞迁移率、穿透基底膜的细胞数量降低,差异均有统计学意义(P<0.05)。结论β-榄香烯能够抑制结肠癌细胞的迁移与侵袭,其机制与β-榄香烯抑制PI3K/Akt信号通路,从而导致Afadin蛋白磷酸化水平下调有关。
Objective To investigate the effect ofβ-elemene on the migration and invasion of SW480 cells,explore the role played by the phosphorylation level of Afadin protein.Methods From October 2019 to January 2020,experiments were performed in the research center of Hebei University of Traditional Chinese Medicine.SW480 cells were divided into control group,IGF-1 group,LY294002 group,β-elemene group,β-elemene with IGF-1 group andβ-elemene with LY294002 group.IGF-1,LY294002 andβ-elemene were used to treat SW480 cells.The expression of Akt,p-Akt,Afadin and p-Afadin was examined by Western blot.The wound healing assay was used to detect the change of cell migration ability,and the transwell invasion test was used to detect the changes of cell invasion ability.Results Compared with the control group,the expression levels of phosphorylated Akt and p-Afadin were increased in IGF-1 group,but decreased in LY294002 group and elemene group;Compared with elemene group,the expression levels of p-Akt and p-Afadin in elemene+IGF-1 group were increased,while those in elemene+LY294002 group were decreased(P<0.05);There was no significant difference in the protein expression of Akt and Afadin among the groups(P>0.05).Compared with the control group,the cell mobility and the number of cells penetrating the basement membrane increased in IGF-1 group,while the cell mobility and the number of cells penetrating the basement membrane decreased in LY294002 group and elemene group;Compared with the elemene group,the cell migration rate and the number of cells penetrating the basement membrane in the elemene+IGF-1 group were increased,while the cell migration rate and the number of cells penetrating the basement membrane in the elemene+LY294002 group were decreased,and the differences were statistically significant(P<0.05).Conclusionβ-elemene can inhibit the migration and invasion of SW480 cells,the mechanism is related toβ-elemene inhibits PI3K/Akt signaling pathway,which leads to down-regulation of Afadin protein phosphorylation.
