摘要
目的观察榄香烯乳对人大肠癌细胞迁移、侵袭的影响,并探讨其可能机制。方法采用不同浓度的榄香烯乳处理人大肠癌HCT116细胞后,以划痕试验方法检测癌细胞迁移能力,以Transwell方法检测癌细胞侵袭能力,采用荧光实时定量PCR方法检测癌细胞miR-155 mRNA水平。结果人大肠癌HCT116细胞经榄香烯乳处理后,迁移和侵袭力均明显下降,且呈剂量依赖性(P<0.05)。榄香烯乳处理组miR-155 mRNA水平明显下调,且呈浓度依赖性。结论榄香烯乳可降低人大肠癌细胞侵袭能力,下调miR-155 mRNA表达是其重要机制之一。
Objective To study the effects of elemene on invasion of human colorectal cancer cell, and discuss its mechanism. Methods After human colorectal cancer HCT116 cell were treated with different doses of elemene, migration of cancer cell was studied by wound scratch assay, and invasion ability were determined by Transwell assay. The miR-155 mRNA were detected by real time fluorescent quantitative PCR assay. Results Elemene could significantly inhibit both invasion ability and a migration in a dose-dependent manner. The mRNA of miR-155 reduced in cancer cells with elemene in a dose-dependent manner. Conclusion Elemene could inhibit migration and invasion of colorectal carcinoma cell through down-regulating expression of miR-155 mRNA.
