摘要
目的探讨胃癌细胞活性与细胞周期相关蛋白(CAPRIN1)表达的关系.方法通过p EGFP-5X-3质粒转染人胃癌细胞SGC-7901,将上调CAPRIN1的表达设为研究组(转染p GEX-5X-3-CAPRIN1质粒),同时设置阴性对照组(转染p EGFP-5X-3空载体质粒)、空白对照组(常规培养).CCK-8法、Transwell小室检测细胞活性;Western blot法检测p-Akt、Cyclin D1蛋白表达水平.结果转染48 h后,与阴性对照组和空白对照组比较,研究组SGC-7901细胞CAPRIN1mRNA和蛋白表达水平明显升高,差异具有统计学意义(P <0.05);SGC-7901细胞CAPRIN1mRNA和蛋白表达水平在空白对照组与阴性对照组之间比较差异无统计学意义(P> 0.05).培养96 h,SGC-7901细胞增殖水平呈上升趋势,在培养48、72、96 h时,与阴性对照组和空白对照组比较,研究组SGC-7901细胞增殖水平明显升高,差异具有统计学意义(P<0.05).研究组的迁移SGC-7901细胞数为(604.17±58.09)个,明显多于阴性对照组的(316.92±38.78)个、空白对照组的(311.75±40.51)个,差异具有统计学意义(P<0.01).与阴性对照组和空白对照组比较,研究组SGC-7901细胞中p-Akt、Cyclin D1蛋白表达水平明显升高,差异具有统计学意义(P<0.01).结论上调CAPRIN1的表达可明显提升胃癌细胞的增殖、迁移能力,可能与激活Akt信号通路和Cyclin D1蛋白有关.
Objective To investigate the relationship between cell viability and cell cycle associated protein(CAPRIN1)expression in gastric cancer cells.Method Human gastric cancer cells SGC-7901 were transfected with pEGFP-5X-3 plasmid,and the expression of CAPRIN1 was up-regulated(transfected with pGEX-5X-3-CARPIN1).The negative control group(transfected with pEGFP-5X-3 empty vector plasmid)and blank control group(conventional culture)were set up.CCK-8 method and Transwell chamber were used to detect cell viability;Western blot was used to detect the expression of p-Akt and CyclinD1.Results 48 hours after transfection,comparing with the negative control group and blank control group,the expression levels of CAPRIN1mRNA and protein in SGC-7901 cells in the study group significantly increased(P<0.05);The expression levels of CAPRIN1mRNA and protein in SGC-7901 cells had no significant difference between the blank control group and the negative control group(P>0.05).The proliferation level of SGC-7901 cells showed an upward trend within 96 hours.Compared with the negative control group and blank control group,the proliferation level of SGC-7901 cells in the study group was significantly higher than that in the negative control group and blank control group at 48 h,72 h and 96 h(P<0.05).The number of migrated SGC-7901 cells in the study group was(604.17±58.09),which was significantly higher than that in the negative control group(316.92±38.78)and the blank control group(311.75±40.51),the difference was statistically significant(P<0.01).Compared with the negative control group and blank control group,the expression levels of p-Akt and CyclinD1 protein in the study group were significantly higher than those in the negative control group and blank control group(P<0.01).Conclusion Up-regulation of CaprinD1 expression can significantly enhance the proliferation and migration of gastric cancer cells,which may be related to the activation of Akt signaling pathway and CyclinD1 protein.
