摘要
CircRNA是一类缺乏5’-帽和3’-polyA尾的共价环状非编码RNA,由前体RNA下游3’末端与上游5’末端反向剪接(backsplicing)而成,具有稳定、丰富、保守、特异表达等特点,包括外显子circRNA(ecircRNA)、内含子circRNA(ciRNA)、外显子-内含子circRNA(EIciRNA)。在沉寂了一段时间后,随着高通量测序技术的进步,近来大量circRNA被鉴定,重新受到研究者的重视。本文综述了自身免疫疾病中circRNA研究的进展,包括其在诊断及疾病分层中的应用以及可能的致病机制等。
CircRNAs are novel class of covalently circular non coding RNAs without 5’-cap or 3’-polyA tail,derived by downstream 3’end backsplicing to upstream 5’of pre-mRNA,characterized by stable,affluent,conservative and distinct expression.CircRNA is composed by exonic circRNA(ecircRNA),intronic circRNA(ciRNA)and exon-intron circRNA(EIciRNA).CircRNAs are been paid attention to again after decades silence after lots of circRNAs identification by universal application of high throughput sequencing.We here have reviewed the updates of circRNA in autoimmunity,including its role in diagnostic,diseases stratification and mechanism.
