基于exoRBase外泌体数据库结直肠癌竞争性内源RNA网络构建及富集分析 被引量:3

Construction and enrichment analysis of competing endogenous RNA network for colorectal cancer based on the exoRBase database

摘要 目的:基于结直肠癌(CRC)患者和正常人血浆外泌体RNA筛选差异性RNA,构建竞争性内源RNA(ceRNA)网络,并进行富集分析。方法:利用exoRBase数据库中CRC患者和正常人血浆标本外泌体RNA[包括信使RNA(mRNA)、长链非编码RNA(lncRNA)、环状RNA(circRNA)]表达谱,依据ceRNA假说构建CRC发生相关的ceRNA调控网络,并对网络中mRNA进行基因本体论(GO)注释和京都基因与基因组百科全书(KEGG)通路富集分析,探索ceRNA网络的潜在功能。结果:共筛选出125个差异表达的mRNAs(DEmRNAs)、53个差异表达的lncRNAs(DElncRNAs)和81个差异表达的circRNAs(DEcircRNAs),并与其对应的miRNA预测数据构建ceRNA调控网络,再对ceRNA网络的25个共同差异表达mRNA进行富集分析。GO注释富集分析显示,CRC ceRNA网络中的外泌体mRNA与癌症相关通路主要富集在GTP结合蛋白RAN通路。KEGG通路富集分析显示,调控网络中差异表达的mRNAs主要富集于促分裂原活化蛋白激酶(MAPK)和血管内皮生长因子(VEGF)信号通路。结论:本研究确定了CRC发生相关的外泌体RNA并构建了相应的ceRNA调控网络,为进一步研究CRC转移进展提供了分子基础,同时为CRC的早期诊断提供了新型的液体活检生物学标志物。 Objective:To screen differential RNA based on plasma exosomal RNA of patients with colorectal cancer(CRC)and normal people,and to construct a network of competing endogenous RNA(ceRNA),and to perform enrichment analysis.Methods:Based on the ceRNA hypothesis,the CRC related ceRNA regulatory network was constructed by using exoRBase database of CRC patients and normal human plasma samples exosomal RNA[including messenger RNA(mRNA),long non-coding RNA(lncRNA),circular RNA(circRNA)]expression profile,and GO annotation and KEGG pathway enrichment analysis were performed on mRNA in the network to explore the potential functions of the ceRNA network.Results:A total of 125 DEmRNAs,53 DElncRNAs and 81 DEcircRNAs were screened out,and combined with their corresponding miRNAs prediction data to construct a ceRNA regulatory network,and then the enrichment analysis of 25 mRNAs in the ceRNA network was performed.GO enrichment analysis showed that the exosomal mRNA and cancer-related pathways in the CRC ceRNA network were mainly enriched in the GTP-binding protein RAN pathway.KEGG enrichment analysis showed that the DEmRNAs in the regulatory network were mainly enriched in mitogen-activated protein kinase(MAPK)and vascular endothelial growth factor(VEGF)signaling pathways.Conclusion:This study identified the exosomal RNA related to the occurrence of CRC and constructed the corresponding ceRNA regulatory network,which provides a molecular basis for further research on the metastasis and progress of CRC,and provides a new type of liquid biopsy biological marker for the early diagnosis of CRC.
