HPLC法同步测定莓茶中二氢杨梅素、杨梅苷、杨梅素含量 被引量:5

Synchronous Detection of Dihydromyricetin,Myricitrin and Myricetin Contents in Mei-tea (Ampelopsis grossedentata) by HPLC

摘要 通过标准品定性、高效液相色谱仪外标法定量、线性关系考察及方法学评价等过程建立了基于HPLC的二氢杨梅素、杨梅苷、杨梅素3种黄酮单体的同步测定方法。结果表明:二氢杨梅素在62~620μg/mL范围呈良好的线性关系,相关系数0.9998,精密度、重现性和稳定性RSD均小于5%,加标回收率在90%~105%;杨梅苷在7.5~75μg/mL范围呈良好的线性关系,相关系数0.9999,精密度、重现性和稳定性RSD均小于5%,加标回收率在90%~105%;杨梅素在6.9~69μg/mL范围呈良好的线性关系,相关系数0.9999,精密度、重现性和稳定性RSD均小于5%,加标回收率在90%~105%;用此法检测12批次莓茶代用茶样品,发现样品中二氢杨梅素含量为249.864~337.551 g/kg;杨梅苷和杨梅素含量分别为2.9815~9.9199和0.4595~3.3677 g/kg。 Through the reference standard qualification,HPLC external standard quantitative analysis,linear relation investigation and evaluation of methodology,the synchronous detection method for three kinds of flavonoid monomers,dihydromyricetin,myricitrin and myricetin,in Mei-tea(a tea substitute:roasted Ampelopsis grossedentata leaves)was established.Results demonstrated that dihydromyricetin displayed a good linear relationship in the range of 62-620μg/mL(R^(2)=0.9998);the RSD of precision,reproducibility and stability each was less than 5%;the recovery rate of standard addition was between 90%and 105%.Myricitrin showed a good linear relationship in the the range of 7.5-75μg/mL(R^(2)=0.9999);the RSD of precision,reproducibility and stability each was less than 5%;the recovery rate of standard addition was between 90%and 105%.Myricetin showed a good linear relationship in the range of 6.9-69μg/mL(R^(2)=0.9999);the RSD of precision,reproducibility and stability each was less than 5%;the recovery rate of standard addition was between 90%and 105%.This method was used to detect 12 batches of Mei-tea samples in which dihydromyricetin content was 249.864-337.551 g/kg,myricitrin and myricetin contents was 2.9815-9.9199 and 0.4595-3.3677 g/kg,respectively.
