摘要
建立了高精度定量毛细管电泳同时测定色甘酸钠滴眼液中色甘酸钠和羟苯乙酯含量的方法。使用带有电隔离槽的石英毛细管柱(总长65 cm,有效长45 cm,内径75μm),以15 mmol/L硼砂-硼酸缓冲液(pH 9.0)为分离缓冲溶液,操作电压-15 kV,检测波长210 nm,恒温20℃。结果表明,各分析物之间均得到良好分离,色甘酸钠和羟苯乙酯峰面积标准偏差(RSD)均小于1.9%。色甘酸钠和羟苯乙酯分别在0.5~5.5 mg/mL,0.01~0.51 mg/mL范围内与峰面积线性关系良好,相关系数(r)分别为0.9929和0.9983,检出限分别为1.3和2.3μg/mL,回收率均在93.1%~100.6%范围内。该方法已用于色甘酸钠滴眼液中色甘酸钠和羟苯乙酯的同时测定。
A capillary zone electrophoresis method for simultaneous determination of cromolyn sodium and ethyl paraben in cromolyn sodium eye drops was established with high-precision quantitative capillary electrophoresis instrument. A quartz capillary( total length 65 cm,effective length 45 cm,inner diameter 75 μm) with an electrical isolation tank was used. The running buffer solution was 15 mmol/L borax-boric acid( pH 9. 0),while the separation voltage was-15 kV,the detection wavelength was 210 nm and the column temperature was20℃,respectively. The results showed that the cromolyn sodium and ethyl paraben could be well separated,and the standard deviations( RSDs) of the peak areas of cromolyn sodium and ethyl paraben were both less than1. 9%. The correlation coefficients( r) of the linear relationships between the concentration and the peak area of sodium cromoglycate( 0. 5-5. 5 mg/m L) and ethyl paraben( 0. 01-0. 51 mg/m L) were 0. 9929 and 0. 9983,and the limits were 1. 3 and 2. 3 μg/m L,respectively. The recoveries were all in the range of 93. 1% to100. 6%. The method has been applied to determine cromolyn sodium and ethyl paraben in cromolyn sodium eye drops simultaneously.
