摘要
目的:探讨止呕方对铂类化疗所致呕吐Wistar模型大鼠血清及脑干5-羟色胺(5-hydroxytryptamine,5-HT)的影响,对中药止呕方的作用机制进行初步探讨。方法:将40只Wistar大鼠随机等分为空白组、模型组(予0.9%的氯化钠溶液)、西药组(予昂丹司琼)和中药组(予止呕方),观察顺铂腹腔注射后各组大鼠72 h内高岭土及正常饲料摄入量;72 h后,处死所有大鼠,采用酶联免疫吸附剂测定法检测大鼠血清、脑干组织中5-HT含量。结果:造模前各组大鼠高岭土及饲料摄入量差异无统计学意义(P>0.05),造模后模型组、西药组、中药组大鼠高岭土摄入量显著增加(P<0.01),而饲料摄入量显著减少(P<0.01),这种情况在0~24 h时最为明显;比较各组大鼠血清及脑干5-HT含量发现,中药止呕方可以显著降低血清与脑干中5-HT含量(P<0.05),其效力与昂丹司琼相当。结论:中药止呕方可以明显改善Wistar呕吐模型大鼠的异食及食欲症状,其中机制可能与降低血清、脑干中5-HT浓度有关。
Objective:To investigate the effects of the Zhiou prescription(止呕方)on 5-HT in serum and brainstem of Wistar model rats with emesis induced by platinum-based chemotherapy,and explore the mechanism of the Zhiou prescription.Methods:40 Wistar rats were randomly and equally divided into the blank group,the model group(took saline),the Western medicine group(received ondansetron)and the TCM medicine group(took the Zhiou prescription).The intake of kaolin and normal feed by the rats in each group within 72 hours after intraperitoneal injection of cisplatin was observed and recorded.Then,after 72 hours,all the rats were sacrificed,and their serum and brainstem 5-HT levels were determined by ELISA.Results:Before modeling,there was no statistically significant difference in the intake of kaolin and feed in each group(P>0.05).However,after modeling,the intake of kaolin by the rats in the model group,the Western medicine group and the TCM medicine group was increased significantly(P<0.01),while the intake of feed was significantly reduced(P<0.01),which was most obvious at 0~24 h.Comparing the serum and brainstem 5-HT levels of rats in each group,it was found that the Zhi’ou prescription can significantly decrease the serum and brainstem 5-HT levels(P<0.05),and its efficacy was equivalent to that of ondansetron.Conclusion:The Zhiou prescription can significantly ameliorate the parorexia and anorexia symptoms in Wistar rats,of which the mechanism may be related to the reduction of 5-HT levels in serum and brainstem.
