天麻素对新生大鼠缺血缺氧脑损伤中小胶质细胞NOX2表达的影响 被引量:9

摘要 目的:研究天麻素(GAS)对新生大鼠缺血缺氧性脑损伤后脑内激活的小胶质细胞表达NADPH氧化酶2(NOX2)的影响。方法:将39只3 d龄新生SD大鼠随机分为假手术组(sham)、缺血缺氧模型组(HIBD)、以及HIBD与GAS联合处理组(HIBD+GAS)。利用左侧颈总动脉结扎结合缺氧箱(8%O_(2),92%N_(2))缺氧2 h制备HIBD模型。采用Western Blot和免疫荧光双标染色检测大鼠左侧胼胝体区NOX2的表达。结果:与sham组相比,HIBD组大鼠胼胝体区NOX2表达明显增高(P<0.05),并且主要表达在活化的小胶质细胞;GAS处理后HIBD大鼠胼胝体区小胶质细胞活化减弱,同时NOX2表达降低(P<0.05)。结论:GAS可能通过降低新生大鼠脑内小胶质细胞NOX2的表达,发挥其神经保护作用。 Objective:To study the effects of gastrodin(GAS)on the expression of NADPH oxidase 2(NOX2)in activated microglia of hypoxic-ischemic brain damage neonatal rats.Methods:Thirty-nine 3-day old SD rats were randomly divided into sham group(sham),hypoxic-ischemic brain damage model group(HIBD)and GAS treatment group(HIBD+GAS).The model is established by ligating the left carotid artery combined with hypoxia(8%O_(2),92%N_(2))for 2 h.The expression of NOX2 in the left corpus callosum of rats was detected by Western Blot and immunofluorescence double staining.Results:Compared with the sham group,the expression of NOX2 in the corpus callosum of HIBD rats was significantly increased(P<0.05),and it was mainly expressed in activated microglia cells.After GAS treatment,microglial activation and NOX2 expression decreased in the corpus callosum of HIBD rats(P<0.05).Conclusion:GAS may exert its neuroprotective effects by reducing the expression of NOX2 in microglia of newborn rats.
