摘要
目的采用网络药理学方法筛选丹参治疗急性一氧化碳中毒迟发性脑病(DEACMP)的作用靶点及相关信号通路,明确其作用机制.方法通过中药系统药理学数据库和分析平台(TCMSP)等数据库搜索丹参的活性成分及靶点,使用GeneCards数据库、DrugBank数据库及人类孟德尔遗传数据库(OMIM)获取DEACMP的靶点,通过蛋白质相互作用检索数据库(STRING)构建蛋白质相互作用(PPI)网络,利用Cytoscape 3.7.2软件构建药物-化合物-作用靶点-疾病网络,应用Metascape数据库进行基因本体(GO)富集分析和京都基因和基因组数据库(KEGG)通路分析.结果检索出丹参的化学成分202个,作用靶点137个,其中与DEACMP有关的作用靶点71个,关键靶点13个.丹参治疗DEACMP可能的机制主要是参与对有毒物质、无机物、活性氧的反应等生物过程,相关信号通路涉及激素代谢、炎症反应、细胞凋亡等.结论本研究通过数据检索明确了丹参具有多个活性成分,能通过多个靶点、多条通路发挥治疗DEACMP的作用.
Objective To screen the Danshen action target spots and related signal pathway in the treatment of delayed encephalopathy after carbon monoxide poisoning(DEACMP)and determine its action mechanism by adopting network pharmacological method.Methods The active components and target spots of Danshen were searched by using the databases of Traditional Chinese Medicine Systems Pharmacology and Analysis Platform(TCMSP),etc.,and the target spots of DEACMP were obtained by applying GeneCards database,Drugbank database and Human Mendelian Inheritance(OMIM)database,the construction of protein-protein interaction(PPI)network was via PPI retrieve database(STRING),the drug-compound-action-target-disease network was constructed by using Cytoscape 3.7.2 software,and the Metascape database was used to carry out gene ontology(GO)enrichment analysis and Kyoto Encyclopedia Gene and Genom(KEGG)pathway analysis.Results A total of 202 chemical constituents of Danshen were retrieved,137 action targets were found,among which 71 action targets were related to DEACMP and 13 key targets.The possible mechanism of Danshen in treating DEACMP is mainly involved in biological processes such as the reactions to toxic substances,inorganic substances,active oxygen,etc.and the related signal pathways concerning hormone metabolism,inflammatory reaction,apoptosis and so on.Conclusions In this study,via database retrieval,the pharmacological mechanism of Danshen in treating DEACMP is preliminarily verified.Danshen clearly possesses many active components and can play a therapeutic role through many target points and pathways for treatment of DEACMP.
