摘要
以平菇菌种作为菌源,经过菌种斜面活化后,对平菇菌丝进行不同的茶浓度培养基培养。结果表明:最适宜的茶汁驯化及种子液培养基为马铃薯200 g、蔗糖20 g、燕麦30 g、磷酸二氢钾2 g、七水硫酸镁1 g、维生素B10.0 1 g、茶叶30 g、水1 L、pH自然。采用单因素试验,以菌丝体干重、菌球个数、粗多糖含量为主要评价指标,来确定培养基最佳的碳源、氮源、无机盐。结果表明:最佳碳源为蔗糖、最佳氮源为酵母膏、最佳无机盐为磷酸二氢钾和氯化钙。基于单因素实验,以菌丝体干重、粗多糖、茶多酚含量为评价指标,采用正交试验方法对平菇菌丝体液体发酵茶培养基进行优化。结果表明:最适宜液体发酵培养基为蔗糖40 g/L、酵母膏6 g/L、磷酸二氢钾2.5 g/L、氯化钙0.5 g/L、茶浓度30 g/L,菌丝体干重10.473 g/L、粗多糖0.19 g/L、茶多酚5.558 g/L。
The Pleurotus ostreatus mycelia from an agar slant culture were cultured in different tea concentrations media.The results showed that the optimal of domestication and liquid seed culture medium was as follows:potato 200 g,sucrose 20 g,oat 30 g,potassium dihydrogen phosphate 2 g,magnesium sulfate heptahydrate 1 g,vitamin B10.01 g,tea 30 g,water 1 L(pH natural).Mycelium dry weight,the number of bacteria balls and the amount of crude polysaccharide were used as the main evaluation indicator,and single-factor test was applied to the selection of carbon source,nitrogen source and inorganic salt.The results showed that the best carbon source was sucrose,the best nitrogen source was yeast extract,and the best inorganic salt was potassium dihydrogen phosphate and calcium chloride.Based on the single-factor tests,mycelium dry weight,tea polyphenols and the amount of crude polysaccharide were used as the evaluation indicators,and orthogonal method was applied to the optimization of liquid fermentation culture medium for Pleurotus ostreatus mycelium.The results showed that the optimal of liquid fermentation culture medium was as follows:sucrose 40 g/L,yeast extract 6 g/L,potassium dihydrogen phosphate 2.5 g/L,calcium chloride 0.5 g/L,tea concentration 30 g/L,mycelium dry weight 10.473 g/L,the amount of crude polysaccharide 0.19 g/L and tea polyphenols 5.558 g/L.
