玉米芯发酵料水浸提液促平菇菌丝生长的机理研究 被引量:6

摘要 以平菇菌丝为试材,研究不同发酵时间(2、4、6、8、10 d)、不同体积分数(20%、40%、60%、80%、100%)的玉米芯培养料水浸提液对平菇菌丝长势、生物量、细胞形态、代谢相关酶(三磷酸腺苷酶、琥珀酸脱氢酶、碱性磷酸酶)和胞外酶(羧甲基纤维素酶、木聚糖酶、漆酶、蛋白酶)活性的影响。结果表明:发酵10 d的培养料浸提液对平菇菌丝生长促进作用最为明显,尤其是体积分数为80%时,平菇菌丝生长速度和生物量分别比对照增加0.53 cm·d^(-1)和1.79 mg,生长速度与100%体积分数无差异,生物量增加2.65 mg;三磷酸腺苷酶、琥珀酸脱氢酶、碱性磷酸酶活性分别比对照高2.6、6.4和4825 U·g^(-1);漆酶、蛋白酶活性分别比对照高11.73、47.6 U·min^(-1)·mL^(-1);此外,三磷酸腺苷酶、琥珀酸脱氢酶、碱性磷酸酶、漆酶、蛋白酶分别比100%体积分数时高5、7.2、2201 U·g^(-1)、2.0 U·min^(-1)·mL^(-1)及14.2 U·min^(-1)·mL^(-1),羧甲基纤维素酶增加了13.09 U·min^(-1)·mL^(-1)。此外,透射电子显微镜观察发现,80%体积分数平菇菌丝细胞内线粒体及囊状体数量较对照明显增加。发酵培养料水浸提液能够明显促进平菇菌丝的生长。 Taking the mycelium of Pleurotus ostreatus as testing material,the effects of aqueous extracts of corn cob medium with different fermentation time(2,4,6,8 and 10 d)and volume fraction(20%,40%,60%,80%and 100%)on the mycelial growth,biomass,cellular morphology,the activities of enzymes related to metabolism(adenosine triphosphatase,succinodehydrogenase,alkaline phosphatase)and extracellular enzymes(carboxymethyl cellulose enzyme,xylanase,laccase protease)were studied.Experimental results show that:the growth promotion of P.ostreatus mycelium was the most obvious after culturing 10 d with corn cob material extract,especially when the volume fraction was 80%.Compared with the control,the growth rate and biomass of P.ostreatus mycelia were increased by 0.53 cm·d^(-1)and 1.79 mg,respectively,however,there was no difference between the growth rate and was 100%volume fraction.The biomass was increased by 2.65 mg;the activities of adenosine triphosphatase,succinodehydrogenase,alkaline phosphatase was 2.6,6.4 and 4825 U·g^(-1) higher than that of control;the laccase and protease activity was 11.73,47.6 U·min^(-1)·mL^(-1) higher than that of control,respectively;In addition,adenosine triphosphate,succinodehydrogenase,alkaline phosphatase,laccase,and protease was 5,7.2,2201 U·g^(-1),2.0 U·min^(-1)·mL^(-1) and 14.2 U·min^(-1)·mL^(-1) higher than 100%volume fraction,respectively,and carboxymethyl cellulase was increased by 13.09 U·min^(-1)·mL^(-1).In addition,it was found that the number of mitochondria and cysts in the mycelia of P.ostreatus with 80%volume fraction was significantly increased compared with the control transmission electron microscopy.The water extract of fermentation medium could significantly promote the growth of P.ostreatus mycelia.
