摘要
目的建立适用于对培植牛黄进行质量评价与控制的HPLC-ELSD测定其中主要胆汁酸类成分的方法。方法使用Agilent SB-C18(4.6 mm×250 mm,5μm)色谱柱;柱温为35℃,乙腈与10 mmol·L^(-1)乙酸铵水溶液为流动相进行梯度洗脱,流速为1.0 mL·min^(-1);ELSD漂移管温度为105℃,氮气流速为1.8 L·min^(-1)。结果胆酸和去氧胆酸分别在0.612~6.120μg和0.425~4.248μg质量范围内进样量对数值和峰面积对数值呈线性关系;胆酸和去氧胆酸的平均回收率分别为103.3%和96.5%,RSD分别为1.1%和2.7%。结论所建方法简单准确,适用于培植牛黄的质量评价与控制,对牛黄类药材的研究也具有一定的参考意义。
OBJECTIVE To establish an HPLC-ELSD method for simultaneous determination of the major bile acids in cultured cow bezoar.METHODS The chromatographic separation was performed on Agilent SB-C18 column(4.6 mm×250 mm,5μm)at 35℃using acetonitrile and 10 mmol·L^(-1) ammonium acetate solution as the mobile phase.Gradient elution was carried out at a flow rate of 1.0 mL·min^(-1).The drift tube of ELSD was maintained at 105℃and the flow rate of nitrogen was 1.8 L·min^(-1).RESULTS The standard curves of cholic acid and deoxycholic acid had good linear relationship in the ranges of 0.612-6.120μg and 0.425-4.248μg,respectively.CONCLUSION The simple and accurate method can be used for the quality assessment and quality control of cultured cow bezoar.The research has reference value for the study on cow bezoar and its substitutes.
